In this study, D6D gene cloned from Oenothera biennis in our laboratory was transferred into the flax genome by Agrobacterium tumefaciens AGL1. And this study can lay the foundation to the transgenic flax that can increase y-linolenic acid. The main results in this study are as following:First, the tissue culture regeneration system of flax has been studied and optimized systemically. By selecting and optimizing factors effected on plant regeneration such as different explants (hypocotyl and cotyledonary node),different combination and concentration of exogenous hormone, the results show that the most favorable explants for flax plant regeneration are hypocotyl and cotyledonary node.Among different combination and concentration of exogenous hormone, 0.2mg/L IAA+0.5 mg/L 6-BA has the highest shoot induction rate and 0.03mg/L IBA+0.07 mg/L NAA is the best for rooting from shoot. Second, the transformation system mediated by Agrobacterium tumefaciens had been studied and optimized systemically based on the flax tissue culture regeneration system which has been established previously. By optimizing diverse factors effected on plant transformation, the results are concluded that:the optimum pro-cultivated time and co-cultivated time of explant hypocotyl and cotyledonary node is 2 days and 0 day,2 days and 5 days respectively. The best concentration of infection Agrobacterium tumefacients of hypocotyl and cotyledonary node is OD600=0.5~0.6,0.8~1.5 and the best time for infection is 5 mins,30 mins respectively.Third, after PCR detection of transgenic regenerated flax, the result indicate that D6D gene was integrated into genome of flax.
