In this study, we took fomentarius from Maoer Mountain, Changbai Mountain, Liangshui and Benxi four areas as targeted bacteria, by length measurement and comparison of mycelium dry weight in the solid medium and biomass changes in the liquid medium, growth rate and enzyme activity related to lignin degradation were detected, compared to the regional Fomes fomentarius growth and physiological characteristics. At the same time, using the relevant sequence amplified polymorphism (SRAP) detection,we compared the genetic diversity among strains of different locations, the degradation capacity of populations for the further study on the wood rot fungi and lignin enzymes and laid the foundation for effective use. The results show that:1. Growth comparison of four regional Fomes fomentariusIn the PDA solid medium, and the four strains growing in the same uniform temperature. The average growth rate of strains in the four regions under 23℃was 0.9cm/d, average growth rate at 28℃was 0.64cm/d. Fomes fomentarius in the growth rate of 23℃is higher than 28℃, which illustrate the growth of Fomes fomentarius is sensitive to temperature. In the same incubation temperature, Fomes fomentarius at different regions had no significant difference in growth rate; the growth rate of the Fomes fomentarius in the same area strain at different temperatures has a significant difference., and the four strains of the Fomes fomentarius under PDA liquid medium has a consistent growth rate at 23℃, growth in different parts of the Fomes fomentarius was no significant difference.2. Comparison of lignin-degrading enzyme activityComparison the activity of the laccase, lignin peroxidase, manganese peroxidase of Fomes fomentarius in four regions, we can see the activity of three types of lignin enzymes in cultured 12 days increasing gradually as time went by. When cultured in different media, enzyme activity had significantly different, due to there were more complete nutrition in PDA medium than in complete medium, and therefore the activity of t lignin-degrading enzymes in the PDA medium were much higher. And Fomes fomentarius in different regions had no significant difference in growth.3.SRAP markers of genetic diversity among populations Fomes fomentariusIn this study we use samples from Changbai Mountain, Liangshui, Benxi, Maoer Mountain (each region 5 samples) as a template in SRAP-PCR system, then selected 10 pairs of combinations from 8×16 couple primers.We used POPGENE32 software to analysis. the electrophoresis results.and calculated the percentage of polymorphic loci. Then we obtained the average percentage of polymorphic loci between four locations was 12.74%, while the average percentage of polymorphic loci in four locations was 6.85% which was lower than the former, so there is greater genetic variation in different places By Nei's index and Shannon index, these two kinds of estimates shows that genetic variation of fomentarius in the various regions hasve the same results:, both showed the lowest diversity index of Changbai Mountain, while the highest diversity index of Benxi. We analyzed the dendrograms between lignin-degrading enzyme activities and genes, we found the results were not completely consistent, but still had relevance.
