| Water-soluble polysaccharide and alkali-soluble polysaccharide were extracted from poria cocos Wolf by different methods, and several important influencing factors to extraction were discussed. A method suitable for extraction was found. This method could enhance the efficiency of extraction to 2.4%, which is 85% higher than the traditional methods.The particle size, the average radius of gyration, and the second Virial coefficient of the pachymaran were measured by static and dynamic laser light scattering (LLS).Sulfate, ferulate, carboxymethyl of pachymaran were studied with the orthogonal design and several affecting factors and levels were discussed in the esterifiable process. Some products were prepared in the optimum condition, which had some changes in the physical properties and the physiological activity. Pachymaran sulfate, pachymaran ferulate, carboxymethyl pachymaran were investigated by IR spectrum.Moreover some special substituent's peak could be found after chemical modification.At the same time, the author also observed the sensitive effects of the sulfate pachymaran associated with cisplatin on 795 human lung adenocarcinoma cells. MTT bioassay was used to test the growth of the tumor cells. Results from MTT showed that the pachymaran had some effect on tumor cells and it could enhance the cytotoxicity of cisplatin on tumor cells when it associated with cisplatin. |
