The generation of microbial enzyme was the first step and also the crucial step in the producing acrylamide using biotransformation method, owing to NHase's activity directly influencing the production efficiency of acrylamide. The fermentation conditions optimization of Nhase and the application of rest cells in preparing acrylamide, using the microbe of Nocardia sp. HD9611, were studied. The results included the following two aspects.I . The studies of fermentation optimizing control of Nocardia sp.HD9611.Studies of growth feature and physiological biochemical characters to Nocardia sp.HD9611.Morphological and the physiological biochemical methods were used to identify the microorganism classfication and appraisal. The results showed that the microorganism belong to I group, Nocardia. It was a mutant of amidase and nitrilase, which could produce and catalyze nitrile compound group to generate amide compound.Researches of the supper-inducing conditions of NHase about Nocardia sp.HD9611.Some potential create induction, such as metal ion, amino acid, vitamin, cell growth hormone, substrate analog and product analog were screened. The results showed that the Co2+ and the urea played a crucial role for action and creation of NHase. It was discovered that INDUCER-FA and INDUCER-AN had strong induction for production of NHase, and the enzyme activities were increased 49.4% and 104.3% respectively. The stage addition mode of inducer was adopted not only can effectively relieve inhibition but also was helpful for enzyme expresses.The growth kinetics of Nocardia sp.HD9611 was studied.Used the automatic controlled BioFLO4500 model number fermentation tank, sugarconcentration, cell density and enzyme activity were determined in properly time using 40% glucose to control sugar concentration in zymotic fluid. The results showed that the um and the Ï„d of Nocardia sp .HD9611 were 0.0859 h-1 and 8.07h respectively. They were raised 120.8% and 107.6% respectively when the total sugar was 80g L-1 in liquid.Researches of catalytic dynamics and de-activity dynamics of Nocardia sp. HD9611.The enzyme is a important factor to affect the yield of acrylamide. So it is important that to produce NHase which had high enzyme activity and maintain its stability. Some kinetic parameters, in which catalytic reaction of enzyme were studied in this essay. The conclusions indicated that Enzyme activity was restrained when the nitrile concentration was greater than 3%. Reaction rate positives correlation with enzyme when the substrate is over-dose .Its fitted linear regression line following: vmax=412.8+351.1+(2958.9+ 57.9)Vcell. The michaelis constant was 0.11mol L-1.Raised temperature could increase reaction speed, on the other hand, devitalized speed would run up when increasing temperature. Correlating the temperature and reaction speed, the activation energy of the hydration of NHase was to be 50.6 KJ mol-1, and the activation energy of thede-active reaction was found to be 12.65KJ mol-1.II .The application of rest cells of Nocardia sp.HD9611 in preparing acrylamide.Now immobilization cell method is extensively adopted in using biotransformation method to produce acrylamide.
