With the development of proteomic and genomics, the technology of molecular bionics has become the topic of biologic study in world. The biomimetic ligands, instead of the natural antibody, can simulate the interaction between antigen and antibody of organism. It has great values for the rapid purifying of proteins, the development of proteomic and the diagnosis of disease.In this paper, a new technology for synthesizing biomimetic ligands will be studied. At the base of simulating the interaction between protein A and IgG, a novel biomimetic ligand of protein A was screened and synthesized by the combination of molecular docking and exsitu-synthesizing.Then, the inreaction between the biomimetic lgand and IgG, and the selectivity of this ligand were assessed using affinity chromatography.The data of experiments indicated that: (1) we screened a series of compounds using molecular docking by which a library of ligands against IgG was generated, then, a novel specific biomimetic ligand N-cbz-L-Tyr were obtained through experimental screening technique.(2) Then, N-cbz-L-Tyr was immobilized on agarose beads to create a specific affinity adsorbent for immunoglobulin G (IgG). IgG with a high purity was purified from human plasma using this new affinity adsorbent. The percentage of IgG in all eluted proteins was 31.2%. (3)A dendritic affinity ligand (Artifical Protein A)was exsitu-synthesized for combining two biomimetic ligand N-cbz-L-Tyr and N-cbz-L-Asp with Gly-L-Gln as the agency. IgG was purified from human plasma using this new affinity adsorbent. The percentage of IgG in all eluted proteins was 77.7%, so the specifity of the adsorbent has been enhanced obviously.Thus, the combination of molecular docking and experimental screening techniques is an ideal approach for discovering specific biomimetic ligands. More than this, the dendritic affinity ligand combining two biomimetic ligand which was synthesized using exsitu-synthesizing technology can improve the selectivity of biomimetic ligand.
