| ObjectivetTo prepare carboplatin-loaded BSA microspheres by emulsifing and thermal denaturing methods and analyse some qualities of the microspheres in vitro , such as , morphological characteristics ,the size distribution , drug loading and encapsulation efficiency for carboplatin(CBP) , and the morphological chang of microspheres digested by artificial intestinal juice at different time point.Methods : The microspheres were prepared by w/o emulsion and thermal fixation method and castor oil was oil phase . The ratio of carboplatin: bovine serum albumin(BSA) was 1:3 , 1:4, 1:6 and 1:12 ; and the stirring speed of emulsion and fixation was 300-500 and 800~1000 rpm , respectively . The emulsion time and fixation time was 10 and 50 min. The emulsion temperature and fixation temperature was 30℃ and 90-100℃. Microspheres so prepared were analysed for morphological characteristics , size distribution, drug loading , and.drug releasing in vitro ._The morphological changes of the microspheres was also observed by electoral microscope . Results: The median diameter of the microspheres was 16.1 μm, with a range of 9.2-33.7μm ; The drug entrapment of microspheres, formulated with differentCBP/BSA ratios as 1:12 , 1:4 and 1:3, was 6.18%, 17.38% and 25.02% (n=3). And theencapsulation efficiencies for CBP during preparation were higher than 80%. Thedeficiency of the microspheres caused by artificial intestinal juice at different time pointcould be observed by electoral microscope . However, the experiment of drug releasingwas failed for some reason .Conclusion: The methods of preparation of CBP-BSA-MS can be used successfully,and the character of the microspheres in vitro is satisfing . The experiment of drugreleasing in vitro need further study. |
