| Glycoprotein is a protein that is composed of oligosaccharides and polypeptide chain combined through the covalent bond. Research has revealed its many biological functions, such as adjusting immunity, inhibiting tumour, reducing blood sugar and preventing oxidation and caducity. As a result, it has a promising prospect in developing new special medicine and functional food. In this dissertation, the ultrasonic extraction of glycoprotein from Chinese yam and its purification, function of immune adjustment were investigated, the main conclusions are as follows:1. Studied ultrasonic extraction of glycoprotein from Chinese yam.Revealed the influences of 7 single factors—total extracting time, ultrasonic pulse time, ultrasonic pulse interval, ultrasonic power, water temperature, rotate speed of electromotor, material-water ratio, on yield of glycoprotein.2. Based on the experiment of single factors, total extracting time, ultrasonic power, water temperature, material-water ratio were singled out for orthogonal test. The results of analysis are as follows:â‘ Range analysis indicates that the optimum extracting condition was total extracting time 40 minutes, ultrasonic power 1400W,water temperature 40℃, material-water ratio 20 gram per litre.â‘¡ Variance analysis indicates that the main factor which has distinct influence on yield was material-water ratio, in turn, the other factors was total extracting and time ultrasonic power.â‘¢ The optimum extracting condition was demonstrated and the yield of glycoprotein was 6. 847%.3. Purified the glycoprotein with EDAE-52 Cellulose and SephadexG-75. As a result, two fractions of glycoprotein-a kind of neutral glycoprotein — GLP- â… ' and another kind of acid glycoprotein — GLP-â…¡' ,were collected respectively. Subsequential experiment of thin-layer chromatography (TLC) testified their high purity. The molecule weight of GLP-â… 'and GLP-â…¡' measured by SephadexG-75 were 89,381 Da and 47, 412 Da. The experiment of TLC revealed that the monosaccharide component of GLP- I was L—galactose, D—glucose, D—mannose. By means of infrared spectrum of GLP-â… ' ,the distinct absorbance of glycoprotein were demonstrated.4. By measuring charcoal particle clearance rate and serum hemolysin level,theinfluence of glycoprotein isolated from Chinese yam water-soluble extracts,on the immunity of normal mice was investigated.Compared low and high dose group with control group,the testing results of chest gland and macrophage phagocytic function were both positive.Therefore,based on the appraising principle and testing methods of functional foods issued by the health ministry of the people's republic of China,the conclusion is that such a glycoprotein has a certain effect on immune adjustment. |
