With the development of the research on smoking and health, there is an increasing growth of requirements on the cigarette safety from world anti-smoking organization. Presently, the emphasis of scientific creativity in tobacco industry are focused on the "reducing tar and toxicants in cigarette smoke", especially "reducing toxicants". Only the amount of harmful compounds in cigarette smoke was selectively reduced, the safety of the tobacco consuming can be improved. The scientific "reducing tar and toxicants in cigarette smoke" is supposed to be based on the quality and quantity analysis of chemical componds in cigarette smoke, especially the harmful ingredients and understanding their physiological and biochemical specialties. Polycyclic aromatic hydrocarbons(PAHs) is one of the most important toxicants in cigarette smoke. In this thesis, the related studies on the basis of extensive literature investigation are summarized in four respects as follows: 1. A novel technique for the modification of traditional smoking machine by adding a series of cartridges filled with absorbents to collect polycyclic aromatic hydrocarbons in mainstream cigarette smoke was proposed. The sampling efficiency was enhanced, therefore, the precision of determination of PAHs was improved. Moreover, the method for the pretreatment procedure of PAHs in cigarette smoke was optimized and simplified. 2. A method for the analysis of 15 PAHs in cigarette smoke by GC/MS-SIM has been developed based on the optimized sample pretreatment procedure and applied to determine the 15 PAHs contents in smoke of a famous cigarette brand at home. The linearity of each PAH is good at the range of 20~1000 ng/mL. The limit of quantitation of benzo[k]fluoranthene is 0.32 ng/mL, far lower than that of former publications. 3. A method for the determination of PAHs in mainstream cigarette smoke by High-performance liquid chromatography with fluorescence detection (HPLC-FLD) was studied and was applied to analyze 6 PAHs in smoke of a cigarette brand. The separating conditions and wavelength program for determination of 6 PAHs in cigarette smoke was optimized. It was shown that this method was characteristic of good repeatability and high recovery, especially fit for use in separating PAH isomers. 4. A real-time monitoring technique of benzo[a]pyrene mutagenicity test with a series acoustic/S. typhimurium (TA 98) sensor was proposed. The optimum assay conditions were experimentally determined and the benzo[a]pyrene mutagenicity test process and the method of its microbial detection was studied. There was a linear relationship between frequency shifts and benzo[a]pyrene concentrations (from 1.2 mg/L to 9.6 mg/L ).
