Solid phase extraction (SPE) is a popular technology for the pretreatment of environmental, biological, pharmaceutical samples. It is progressed on the bases of liquid-liquid extraction, the rule of similarity and the characteristicis of the solid phase. It is considered as the standard pretreatment method in environment analysis by Unite State Environment Protect Agency (USEPA). At present, this technique has been used in many fields in china, e.g. the analysis of polynuclear aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs) and some other organic compounds in water; the trace pesticide or herbicide in fruits, vegetables and foods; the antibiotics, the clinic pharmacy and so on.The chromatographic column of SPE which was prepared by the molecularly imprinted technology was used for pretreatment of environment sample. The results showed that the molecularly imprinted polymers exihibted significant molecular recognition ability and high affinity for paclobutrazol in aqueous system, which could be applied for trace analysis of paclobutrazol in water. In addition, a continuous ion-exchange chromatographic column was prepared, by which the drug concentration in plasma sample could be determined. The retention behavior of the polymer had been researched and it could be concluded that the hydrophile and the activity of ion exchange were both obvious on the column. The continous work was carried on. In this paper, three chapters were consisted of.Chapter I: the principle, applications and sorts of SPE were introduced. Also the application of MIT and ion-exchange chromatography were reviewed.Chapter II: Paclobutrazol had been employed as template for the preparation of MIP, which was applied as stationary phase for the separation of paclobutrazol and its analogies. The characteristics of adsorption and thermodynamics of paclobutrazol - MIP were investigated. The adsorption isotherms of paclobutrazol were studied by static method. It was concluded that two types of binding site were present in the imprinted polymers after Schatchard analysis. The results showed that the MIP could be applied as SPE material for trace analysis and enrichment of paclobutrazol in water.
