| Using microbial fermentation community to dispose of livestoke and poultrymanure is the most popular problem now. Cellulose decomposing microorganisms anddeodorizing bacterium, as the most important two parts of the microbial fermentationcommunity, were found to be very useful in the utilization of livestock and poultrymanure. In this paper, a composite microbial system FH3 was gained and itsproperties concerning enzymatic activity were extensively studied. Two strainsdeodorizing bacterium CB5 and CS14 were separated and were composed with FH3into a microbial fermentation community, which was applied into the fermentationexperiment.23 strains of cellulose decomposing microorganisms were isolated in advantage ofcellulose-congo red medium. Among them 4 strains had the higher cellulose enzymeactivity. Through conbination experiment, a composite microbial system FH3 wasgained and its properties of cellulase were studied.Studied upon the effect of culture temperature, medium initial pH, revolution,carbon sources, nitrogen sources and culture time on the enzyme production of FH3.The results showed the optimal enzyme production conditions of FH3. The mediumwith bran and filter paper as carbon and pepotone as nitrogen source was optimal andthe maximum cellulase activity was reached among 30~35℃ and pH 6.0~6.5 whencultured for 48~96hThis paper investigated the effect of temperature and pH on the enzyme reaction ofFH3. The thermal stability of enzyme of FH3 was researched too. The results showedthe optimal conditions of enzyme reaction of FH3. The temperature was among 45~55℃ and pH was among 4.5~5.5. The thermal denaturation was approximately 60℃.15strains of deodorizing bacterium were isolated in advantage of the solution ofmanure. Among them a strain of bacteria CB5 and a strain of actinomycetes CS14showed the high ability of deodorization. Combinated the two deodorizing bacteriumwith FH3 as the microbial fermentation community and used it in fermentationexperiment. Applied the microbial fermentation community into the fermentationexperiment. The results showed that temperature of compost, which was inoculatedmicrobial fermentation community reached the highest temperature at the fouth dayand last a long time at high temperature. C/N and the ratio of crude cellulose weredecreased quickly. The ratio of nutrition such as N, P, K, etc was much higher than thecompost, which was not inoculated microbial fermentation community. |
