The paper used to tomato skin that the ketchup factory dealt with waste material as raw material to study tomato skin's advance treatment and preservation. The results are as follows: To separate tomato skin and seed by water, then dry the skin with spinner, vacuum packing, in room temperature after 10 days lycopene remain rate is 56.1363%. The preservation method can apply to short-term preserve tomato skin.By the comparison of different solvent extractive effect, we determined the optimum extractive solvent is solvent A and the optimum technologic conditions of solvent extraction were as follows: extract of lycopene with 1:8 ratio of raw material to solvent A for 70min under T=40℃,extracted three times, extraction rate can achieve 87.3216%.By researching the lycopene's extraction rate of solvent A and ethyl acetate in different raw materials, be proved that ethyl acetate of lower extraction rate in tomato skin attribute to the affect of the waxiness and water of tomato skin.Microwave extraction's research showed: extraction solvent was solvent A, extraction time was 60s, extraction power was 360W, ratio of liquor and material was ll:l,the number of extraction stage was 4, extraction rate can achieve 93.5285%. If reached the same extraction rate, only by solvent and heating extraction will take 280 min.To observe the tomato skin with microscope: the results showed that treated tomato skin cell had plasmolysis, and cell membrane's osmosis turned stronger, intracellular fluid exuded from cell, these may be one of the reason that microwave can improve the extraction efficiency.The content of lycopene in tomato oleoresin was determined by HPLC. The conditions of HPLC were as follows: the best mobile phase was acetonitrile : dichloromethane : methanol (7:2:7) detected at 472nm. Use C18 of chromatographic column CLC-ODS (150mm × 4.6mm ID, 5 μ m), velocity of flow was 1ml/min, temperature of column was 25℃. The retention time of lycopene was 14-14.6min.And by using TCL and LCC, studied others components of tomato oleoresin. The results are as follows: using a mixture of petroleum ether : solvent A was 19:1,9:1,4:1 as developing agent, analysis with TLC revealed that there are six major ingredients in oleoresin, and the Rf value of lycopene is 0.73. By isolation and purification of LCC, not only getting the purer lycopene but also estimating others components of tomato oleoresin, by UV/Vis absorption spectroscopy tested wavelength and contrasted to literature, estimated preliminary three ingredients in tomato oleoresin may be β -Carotene, Phytoene, Lycoxanthin.
