Monolithic column, a. new type of column, has attracted more and more attention in recent years. There are many advantages over traditional packed columns, such as easy preparation, excellent permeability, and ability in fast and high-selectivity separation. In this study, a series of monolithic columns for high-performance liquid chromatography were prepared by in-situ copolymerization of glycidyl methacrylate (GMA) and ethylene dimethacrylate (EDMA) in the presence of a porogen solvent containing 1-propanol, 1,4-butandiol and water. Then bovine serum album (BSA) was immobilized on the obtained monolithic matrix to be the stationary phase of molecular bio-chromatography. Finally this monolithic column was applied in analyzing the methanol extraction of traditional Chinese medicine, Angelica Sinensis (Qliv.) Diels. The ratio of porogen solvent to total monomer in the polymerization mixture and the composition of porogen solvent (1-propanol, 1,4-butandiol...
