Resveratrol , a kind of phytoalexin, has many biological and pharmacological activities, such as anti-cancer, anti-oxidant, anti-mutagenic, anti-microbial and anti-microbe and anti-inflammatory. It can protect against atherosclerosis and coronary heart disease. It is also benefit to cardio-vascular. Therefore, resveratrol will surely has a broad application prospect.Polygonum Cuspidtum Sieb. Et. Zucc belongs to the family of pol ygonaceae, The theoretic content of resveratrol in cuspidat urn is the highest among all studied plant species a ccording to known. So the purification and determination of resverat rol from cuspidtum becomes a research focus.1. The thesis offers a premininary analysis on separation condition of resveratrol from cuspidatum. The component change in sample after extracted with chloroform, and Ethyl Acetate was compared; Through comparison of different eluant, various eluant concentration and kinds of flow rate, find the best solution of resveratrol in LH-20 chromatogram. Optimum purification condition is elute with 60% methanol with a flow rate of 0.5ml/min . After LH-20 Chromatogram elution, the purity of resveratrol was determined by MPLC to be as high as 80.34% .2. To establish a MPLC method for the content determination of resveratrol in Polygonum cuspidatum Sieb. Et. Zucc, Analytical column SOURCE 5RPC ST 4.6/150 was used with a mixture of acetonitrile+water (30+70,volume ratio ) as mobile phase ,with a flow rate of 1ml/min and the UV detection wavelength was 306nm, The linear range was 0.6-1.6mg/ml (r=0.9919). The average recovery of prodatin was 103.0%-90.4% (RSD=7.21%). The method is convenient and accurate. Result indicated a reverse phase chromatography media based o n polystyrene/divinyl benzene has excellent resolution for resveratrol in cuspidatum. P rovide theoretical support for process development. |
