| [Background]Soluble dietary fiber is significant to physiological function, while proportion of Soluble dietary fiber is very small in raw resource, which cannt reach to the remand of the balance of dietary.Recently, many researchers study on the modification of dietary fiber by diverse methods. We prepared lemon peel dietary fiber by fermentation of Trichoderma Viride, and to explore its toxicological safety assessment as a health food. [Methods]Extraction dietary fiber from lemon peel by fermentation of Tricho-derma Viride, the optimum ferment conditions were determined by orthogonal design test. Acute toxicity test, the LD50 of lemon peel DF was determined Horn's method to KM mice. Micronucleus test of bone marrow cell as well as sperm shape abnormality test in mice, used to detect the genetic toxicity of lemon peel DF. 30 days feeding test, S-D rats, weighting 60-80g, were divided in 4 groups randomly. One group was fed with normal rat feed as Control group, the other three groups 2.5%, 5% and 10% of lemon peel DF in the rat diet. Tail blood of the animals was collected to determine the haematological parameters. Serums were obtained from all of the rats to measure clinical chemical parameters. The rats were anatomized and observed histopathology, and major organs were measured. [Result]The result of orthogonal experiment indicated that the optimum conditions for extraction of dietary fiber from lemon peel by fermentation of Trichoderma Viride were: fermentation temperature 32℃, taking-time 72h, pH6.3, the product of lemon peel dietary fiber, its swelling capacity and water holding capacity were 6.76ml/g, 16.57g/g respectively.Acute toxicity, the oral LD50 values of lemon peel DF for male and female KM mice... |
