Beneficial effects of probiotics realated with survival number and subsisttent quality in intestines, but gastric acid and bile can kill it. It affects the beneficial effects of probiotics. In order to tolerate gastric juice and bile, and cause the probiotics multiplys in the intestines and play the profitable action, this research adopted " Microcapsulation" and " Intestinal-lysis " technique. The technological processes about preparing microcapsulation of LA with complex emulsification method were studied. The sodium alginate was used as shell-material and LA, high efficient protectant that used protect cell were used as core-material. Study on the methods of enteric-coating microcapsules containing LA and the effect element, and researcied the different methods and adding different freeze-drying protectants to prepare LA of microcapsules. Study on the microstructure, diameter, trapping efficiency of LA of microcapsule on this base.and research the distintegrated characteristic microencapsulated L. acidophilus in simulated gastric fluid, bile and simulated intestinal fluid. Researcied the storing characteristic of 4℃and 37℃, the results were as follows:1. Both external emulsification method and internal emulsification method Can prepare LA of microcapsules, survival rate of LA loaded in microcapsules, which prepared by internal emulsification method are higher, the range of Diametre is narrower, the surface are more glossy and have well global form.The results of external emulsification method showed that the optional conditions were as follows: concentration of sodium alginate was 2%, emulsification time was10min, stirring speed was 400rpm, proportion of LA and sodium alginate was 1:6.The results of internal emulsification method showed that the optional conditions were as follows: concentration of sodium alginate was 2%, emulsification time was 10min, stirring speed was 400rpm, proportion of LA and sodium alginate was 1:6.2. If added 40g/L cornstarch, 15% glycerol and 60g/L inulin, viable bacteria can increase to 2.2×1011cfu/g. The modified method increased the survival of bacteria by 1000-fold compared with comparison series.
