Mass growth of cyanobacteria (blue-green algae), leading to the production of blooms, can occur in ertrophic waterbodies throughout the world, the Microcystins pollution has already become a environmental question that the whole world pays close attention to, Microcystins are toxic metabolites released from cyanobacterial bloom, can cause poisoning of animal and also pose a health hazard to human. Require urgently that carries on fast qualitative, quantitative analysis to Microcystins in the drinking water source. Purifed Microcystins are needed to utilize as the standard and reagent material in a range of extensive research applications including biochemical and ecotoxicological studies.The retention behaviors of Microcystins and separation condition on reverse phase HPLC setup were systematically investigated. Optimum HPLC system factor and mechanism were developed. A method for trace level analysis of Microcystins in water using SPE and HPLC has been developed in this report, which is fast, sensitive and suitable for the determination of the two most common Microcystins, MC-LR and MC-RR. This method enables the absolute determination of Microcystins even at the level as low as 1.00ng, and the linear range is from 0.125μg/L to 50μg/L, recovery of two Microcystins is respectively 97.35%(MC-RR), 92.47%( MC-LR). This experiment analyzed the real water sample from Meiliang Bay of Taihu Lake in Wuxi, there are two Microcystins mainly in it, and their content is respectively MC-RR3.348μg/L, MC-LR0.941μg/L.Referring to the methods available of Microcystins extraction and purifying, and ameliorating them, we used blue-green algae in Taihu Lake to prepare MC-LR and MC-RR samples of high purity by using high-performance preparative liquid chromatography. The samples were analyzed by HPLC and the purity exceeds 98%. Two Microcystins standards were gained by evaporating samples, mass of two Microcystins is respectively MC-RR121.1μg and MC-LR62.8μg.
