| This study was based on the recently proposed research pattern of Traditional Chinese Medicine (TCM)– multi-component library (MCL). Danggui (the roots of Angelica Sinensis (Oliv.) Diel) was selected as the research target of the study. A systematic separation method for Danggui was founded on the multi-model separation techniques which would be served as complementarities of others. And some kinds of modern analytical techniques were utilized to elucidate the chemical characterization of the components of Danggui. Sequentially multi-components library and data library were established, which provided demonstration for the systematic separation methods and chemical characterization of other TCMs.A stable and reliable HPLC method was developed for the analysis of the water extract of Danggui, a complex system. The mobile phase, additive and some other aspects were optimized. The HPLC retention parameters a, c were obtained quickly and correctly by using five linear gradients, which could change unknown components into chromatographic known components.The systematic separation method adopted multi-mode separation techniques, including solvent distribution, macroporous absorbing resin and preparative chromatography. 3 components separated from water-solubility components were obtained by macroporous absorbing resin and 36 components separated from ethyl acetate components were purified by preparative HPLC. All these components constitute the multi-component library of Danggui. And they were characterized by optimized HPLC analysis method, which could offer data information of Danggui for the data library.By making use of LC-MS preparative chromatographic technique, 2compounds, ferulic acid and senkyunolide I, and 1 mixture, composed of 2 phthalides, senkyunolide G and 3-hydroxy-butylphthalide were prepared. This preparative technique supply a credible approach for repeatedly obtaining compounds from relatively simple components. And the information of the obtained compounds could provide reliable information for the data library, which would make the process of identifying unknown compounds easier and more reliable.HPLC-MS-MS was used to analyse the main components in Danggui, which made the hypostasis of material base of Danggui clearer. Furthermore, the MS fragmentation regularity of the phthalides was proposed via the MS data information of the 3 prepared compounds. 3 known phthslides and 2 unknown dimeric phthalide derivatives were tentatively identified based on the presumed MS fragmentation regularity of the phthalides, which provide an accurate and fast method to identify the phthalides, and the comparison of their UV spectra with those published in literatures. |
