As a kind of efficient and low poison anti-tumor medicine,β-elemene has got the extensive applied foreground. It was usually used as emulsion. Because phlebitis was often arosed after the emulsion made fromβ-elemene was injected into vein, its use was limited badly. To resolve the problem, we makeβ-elemene intoβ-elemene liposomes utilizing fully the characters of liposomes. Two methods--reverse phase evaporation method and thin-film hydration method were used in the experiments. Regarding envelop ratio as the test aim, we located the best material ratio on those preparing methods by experiments. As the liposomes are prepared by thin-film hydration method, the best weight ratio of lecithin to cholesterin andβ-elemene is 400:34:40.8; as the liposomes are prepared by reverse phase evaporation method, the best weight ratio of lecithin to cholesterin andβ-elemene is 400:51:54. The envelop ratio of them is over 85%. It show thatβ-elemene is proper to be enveloped by liposomes.In order to produce liposomes in the gross, a new method—Reverse Phase & High Pressure Extrusion that was adapted to industrial product was put forward by integrating several different methods of preparing liposomes. Based on it , a new technnology and equipments were designed. The capacity of the system is 5 litres per batch. Utilizing the new equipments, we prepared grease-solubilityβ-elemene liposomes and water-solubility arbutin liposomes. The diameter of theβ-elemene liposomes prepared through the new equipment is 108nm and the envelop ratio is 96.408%. The diameter of the arbutin liposomes prepared through the new equipment is 70nm and the envelop ratio is 16.532%. The results are close to those educed from the laboratory and this illuminate the new craft can satisfy the demand of the actual production in quality.
