The Extraction Technology Of Semen Of Platycladi And Study The Vivid Chemical Compositons

Semen platycladi is the kernel of arborvitae, which is a kind of cypress. Semen Platycladi has the effects of tranquilizing by nourishing the heart and loosening the bowel to relieve constipation. It is effective in treating nervous exhaustion, palpitation, insomnia, constipation and so on in the clinical. It is low toxic, safe using and has very distinctly officinal value.This thesis includes two parts:The first part introduces the herbal medicine origin chemical components, pharmacological effects and clinical applications.The second part includes:(1) The fatty acid composition from the seeds of oriental arborvitae was investigated. Fatty acid methyl ester was analyzed by combining capillary gas chromatography/mass spectrometry.The results show that there are seven chemical compounds. Identify five kinds of fatty acids methyl ester. The main unsaturated fatty acid include 9,12,15-Octadecatrienoic acid and Methyl (Z)-5,ll,14,17-eicosatetraenoate, which make up 82.29% of the total fatty acids.They are with of high nutritional value.(2) Wister second-generation rats are feed with forage, which is added by fatty oil from the semen platycladi. We study the male and female rat's behavior thorough the Y Maze and Morris' water maze .Y maze show that the result of Y Maze is best in the three group. There are not marked difference in these two groups,but there is marked difference between the two groups.In the Morris' water maze experiments,there are not marked difference in these two groups.(3) Feed the Kunming rats with forage introduced by STZ and Alloxan , which is added by fatty oil from the semen platycladi. We study the diabetic rat's cholesterol and blood fat.The result shows that there is a slight decrease in the biood sugarduring the three weeks, but there is a great decline in the later three week. Both oil of semen platycladi and oil of tea have the effect of hypoglycemic and the effect of former is better than later. But it spends more time to take effect.Oil of semen platycladi regulate the lipid metabolism of rats with forage and has marked decrease in the blood fat disturbances caused by high blood sugar.(4) Confirming the best extractive techniques of the total saponines, through the four factors(concentration of ethanol, time of extraction and times, dosage ofextractive solvent and temperature of extraction )The levels orthogonal test, according to the characters of the main saponines of semen platycladi. The best extractive techniques is that adding 10 times 95% ethanol to the medicinal materials, refluxing 3 times,4 hours per time and the refluxing temperature is 60℃ .This method is a good reference for the industrialization. Confirming the structures through the method of chemistry reagents, UV, IR and so on.
