| The apple outputting in China occupy the first place in the world. Artifactitious applewhich processes in the juice high up to ten million ton annually, corresponding to producea great deal of skin residue wastes. The apple seed in the skin residue can not getexploitation at a long time. The constituent of apple seed oil is mainly high unsaturatedfatty acids, which can be directly used for cosmetics and medicine. Additional value ofproduct is very high. Therefore, aimed at the development of new resources—apple seedoil, this paper studied the method of extraction on apple seed oil.(1) Method of solvent extraction on apple seed oilBased on the sieving of normal hexane as the best solvent to apple seed oil ofextraction, this paper studied the influence of factors as extraction time, temperature andliquid-solid The results showed that the influence of all factors was notable, in whichextraction time influenced biggest, while solid—liquid ratio compared secondly. With themethod quadratic-form recursive orthogonality, the paper got the best extractionconditions as follows: treatment time 7.3h, the liquid-solid ratio 3:1 and the temperature30℃.Under the best condition, the max rate of oil extraction reached to 89.48%, and alsogot the extraction model as follows: Y=-438.424+29.06327×t+52.08819×R +77.52558×T-2.040902×t×t-8.387195×R×R 4.339002×T×T(R stands for the liquid-solidratio).The model match highly after square analysis (P失拟>0.05).(2) Method of water extraction on apple seed oilMaking use of the superior condition in single-factor experiment, the paper got thebest technological parameters of water extraction on apple seed oil as follows: the numberof phage 60, extraction temperature 60℃, extraction time 5 h, the aqueous pH 11.0 andthe liquid-solid 5:1.Under this condition, the extraction rate of apple seed oil was 35.5%(3) Method of water-enzyme extraction on apple seed oilDuring the water extraction on apple seed oil, we added protease to the the systemand studied the influence of factors as aqueous pH, extraction temperature and extractiontime on the extraction rate of apple seed oil. The results showed that the influence of allfactors was notable, in which the extraction time influenced biggest, while extraction temperature compared secondly. With the method of quadratic-form recursiveorthogonality, the paper got the best extraction conditions as follows: aqueous pH 9.0,temperature 60℃,extraction time 115.23min.Under the best condition, the max rate of oilextraction reached to 75.44%, and also got the extraction model as follows:Y=-500.4+60.88129×pH+8.920814×T+0.497104×t-3.39348×pH×pH-0.072295×T×T-0.002121×t×t.The model match highly after square analysis (Pr>0.05).(4) Method of enzyme-assistant three phase partitioning (EATPP) on extraction ofapple seed oilCombining the method of solvent extraction, water extraction and water-enzymeextraction, the paper studied enzyme-assistant three phase partitioning on extraction ofapple seed oil. In solution of pH 9.0, added content of alcalase protease 1%(mL/g), waterbath temperature 60℃, extraction time 115.23 min, added content of ammonium sulfate30%(w/v) and normal alkane 1:1(v/v), the extraction rate of apple seed oil was 85.5%.Synthesized the study above, 4 extraction methods had its own merit and shortcoming.The rate of solvent extraction was tallest, reaching to 92.31%; The method of waterextraction was safest, but the efficiency was lowest. According to the accession ofprotease, the extract rate reached to 75.44%. In this foundation, method of EATPP couldimprove the extraction rate to 85.5%, which can improve the extraction rate, reduce thedosage of organic solvent and receive an environmental protection performance. |
