This thesis is mainly dedicated to research in the separation, purification, analysis and quality control of polysacchrides from a widely used Chinese herbal drug, poria cocos or Fu-ling. Fu-Ling, which grows on the roots of Pine trees, is the sclederma of Poria Cocos(Schw.) Wolf. Based on the major pharmacological effets,Fu-Ling is one of the major ingredients of nearly 40 TCMs, which have been used for more than thousands of years in Chinese history. Its polysacchrides have many biological activities, such as anti-tumor.The efficiency of classic extraction method for polysacchrides by hot water was studied. The influence of extraction temperature, water ratio, extraction time and alcohol concentration on the yield of sugars from Fu-ling was inspected in the process of extraction. The optimal condition was chosen for the extraction of polysaccharide from Fu-ling(condition: water ratio 1:9.4, temperature 90℃,time 7 hours,precipating alcohol concentration 70%).In the process of purification for rude sugars,the efficiency of deproteining by sevag method,papain method and sevag method combined with papain was studied. The result showed that sevag method combined with papain was best for isolating protein from sugars.Ultrafiltration method was also studied in separating sugars of different mocular weights.Ultrafiltration method and High-performance Liquid Chromatography were employed for the fingerprint profiling of carbonhydrates from a popular medicinal fungus known as Poria cocos or Fu-ling. The feasibility of employing the High-performance Liquid Chromatography (HPLC) peak profiles of carbohydrates for the screening of the polysaccharides extracted from various Poria cocos samples of different origins(Guangxi,Hunan,Yunan) was demonstrated. The similarity of Yunnan Fu-ling and Hunan Fu-ling in comparison with Guangxi Fu-ling is 0.9190 and 0.7400, respectively. The methodologies described in this thesis should be useful in the quality control of various TCM polysaccharides products.
