Cinnamomum burmannii and Omanthus fragrans are characteristic plant resources of Guangxi. There are abundant and extensive resources in Guilin. A great deal of berrys were fallen off on the ground when it was mature every year. It not only polluted environment but also wasted a precious resources. But research and exploitation about two kinds of berrys has not been reported yet. Exploitation and use of the berrys can obtain a good economic benefit and has a social significance.Part 1: Exploitation and use of berrys of Cinnamomum burmannii.Isolation, purification, physical and chemical properties, structure and bioactivity of melanin extracted from peel of Cinnamomum burmannii'berry included in part one are as follows:Isolation and purification of melanin from peel of Cinnamomum burmannii's berry: the influence of each factor were analyzed and the optimum condition of extracting melanin from Cinnamomum burmannii's berry was the ration of 1 (material): 30 (10% NH3·H2O), room temperature for two times and for 24 h once time, acidified by addition of 2 N HC1 to pH 2.5 for deposition of melanin and filtered to obtain the crude melanin. The crude melanin was hydrolysed with 7 M HCl for 6 h, extracted with petroleum ether and chloroform in turn and purified on a Sephadex G-75 for getting purification melanin.Physical and chemical properties of the melanin from peel of Cinnamomum burmannii's berry: it can be dissolved easily in alkaline aqueous, was stable to light, temperature. Reducer, salt, organic acid and preservative insignificantly affected the melanin. Moreover, melanins are including some little elment such as Fe,Cu,Zn. Acute toxicity test showed that the maximum tolerance dose of rat is more than 12.85 g/kg.Structure analyse of melanin from peel of Cinnamomum burmannii's berry: according to oxidative degradation, UV, Raman spectra, infrared spectra, ESI-MS, X-ray, 13CNMR, 1HNMR spectra and elemental analyse etc, melanins contain DHI and DHICA units, DHI and DHICA were considered as the primary melanin precursors of eumelanins.Biological activity of melanin from peel of Cinnamomum burmannii's berry: according to biological activity test of melanin, it is a avirulence natural pigment and can enhance sun protection factor of sunscreen. For example, SPF of sunscreen samples combination with melanin increased obviously. Moreover, melanin can inhibited the growth of HaCaT cells and has significant antioxidant activities. These results suggest that melanin may be an effective inhibitor of skin pigmentation stimulated by UV radiation and may be valuable in the prevention of UV-induced photo-aging and melanoma.Extracting grease from Cinnamomum burmannii's seed: the optimum condition of extracting grease from Cinnamomum burmannii's seed was as follows: 1 (material): 7 (petroleum ether),50℃for two times and for 40 minutes once time. 1,2,3-tris-lauroyloxy-propane was seperated from the grease for the first time and yeld of it is 40%. Discovered a new method to produce glycerol monolaurate as a preservative in food industry from Cinnamomum burmannii's seed.Part 2: Exploitation and use of berrys of Omanthus fragrans.Isolation, purification, physical and chemical properties, structure and bioactivity of melanin extracted from peel of Omanthus fragrans'berry were studyed in part two.Isolation and purification of melanin: the optimum condition of extracting melanin from peel of Omanthus fragrans'berry was the ration of 1 (material): 30 (10% NH3·H2O), room temperature for two times and for 24 h once time, acidified by addition of 2 M HC1 to pH 2.5 for deposition of melanin and filtered to obtain the crude melanin. The crude melanin was hydrolysed with 7 N HCl for 6 h, extracted with chloroform and purified on a Sephadex G-75 for getting purification melanin.Structure analyse of melanin from peel of Omanthus fragrans'berry: according to oxidative degradation, UV, Raman spectra, infrared spectra, ESI-MS, X-ray, 13CNMR, 1HNMR spectra and elemental analyse etc, melanins contain DHI and DHICA units. But a little different in the structure with melanin from peel of Cinnamomum burmannii's berry.Physical and chemical properties of the melanin from peel of Omanthus fragrans'berry are similar to Cinnamomum burmannii. Acute toxicity test showed that the maximum tolerance dose of rat is more than 10.0 g/kg.The two iridoids are the main component of the seeds of Omanthus fragrans and their content were analyzed by HPLC. That the optimum reap time is the last ten-day of April was determined according to the content of two iridoids and it can provide theory basic for scientific reap and production. |
