Study On Extraction And Preparation Technology Of Extracts From Rhizoma Anemarrhenae

Diabetes is a serious threat to human health in worldwide public health problem, seeking an effective drug for treatment of diabetes has been on the hot spots.In this study,in order to maximize the efficacy of treating diabetic, based on the use of traditional Chinese medicine with modern pharmacological research results, the process for comprehensive extracting the active ingredient is designed. And extract is purified, in order to develope a new type Traditional Chinese Medicine for treating diabetes.In this study uniform design and orthogonal experiment are used, the optimization extraction process is chosed to comprehensively extract the active ingredients for treating diabetic. First, take ethanol as an extraction solvent to extract timosaponin and benzene topiramate ketones. Second, use water to extract anemaran, and enlarge the certification test to prove that the extraction technology is basic stability.In order to concentrate the valid target and achieves the separation purification the goal, the author uses the macroporous resin to purify as the material, chooses the suitable density of extraction solution and the volume, and selects the appropriate ethyl alcohol density and the volume to take the eluent, confirms the best Purification Process for alcohol abstracts ultimately. At the same time, using membrane separation technology, select the appropriate membrane pore size and the best purification process to purify water abstracts, which contain anemaran more than 70 percent.According to the physical and chemical properties of raw materials and the characteristics of tablet,extract of Rhizoma Anemarrhenae is made to tablet. Take hardness and disintegration as the inspection targets, the type and amount of the filler, disintegrating agents, lubricants, and other adjuvant are selected, and the preparation Process is optimized, and a film clothing make the agents more stable and easy to store.Application of ELSD and PDA detector as well as LC/MS and other advanced analytical tools, a preliminary study on the main ingredient in extract of Rhizoma Anemarrhenae is took. The main ingredient in alcohol extract of Rhizoma Anemarrhenae is timosaponin and benzene topiramate ketones.According to the requirements in the appendix of the Chinese Pharmacopoeia 2005 version 2, the quality analysis of the preparation is carried on, including preparation traits, identification, inspection, determination and so on. The content of timosaponin and benzene topiramate ketones is determined by HPLC, while the content of anemaran is determined by UV. The experimental conditions exploration and methodological survey are carried on, this method can determine the exact content of samples. Based on this, the author has formulated the quality specification of the preparation.
