Nitrofuran metabolites and malachite green are two kinds of veterinary. They can enter human body through food and cause harm to human health. Therefore, the detection of nitrofuran metabolites and malachite green residues has been strengthened in all countries of the world.In this paper, on the background of lack of analysis method for the trace veterinary residue in food, two analysis methods for four nitrofuran metabolites and malachite green, such as furazolidone, furaltadone, nitrofurazone, nitrofurantoin in honey and salmon were investigated systematically. The pretreatment method of sample was discussed and the simultaneous detection of four nitrofuran metabolites and malachite green using two LC-MS methods have been established respectively.During the pretreatment procedure, the extraction of four nitrofuran metabolites and malachite green was carried out by liquid-solid and liquid-liquid extraction, and the extract was cleaned up by alumina SPE cartridges. The optimum pretreatment conditions were obtained by optimizing the conditions of extraction and cleaning up. The optimal conditions of solid-phase extraction were also studied. Average recoveries of spiked analytes respectively ranged from 67.0% to 112.2% and from 95.6% to 103.3%, and the repeated relative standard deviation (RSD) for the recoveries ranged from 3.0% to 9.4% and from 4.8% to 6.1% respectively.The conditions of HPLC and MS were developed respectively for simultaneous detection of four nitrofuran metabolites and malachite green in honey and salmon by high performance liquid chromatography-mass spectrometry. Under certain conditions, analytes could be separated well and the repeated RSDs of the mixed criterion solution were 0.5~1.0% and 0.2~0.7%. The limits of detection for the two methods reached at 0.01~0.25ng/g and 0.15~0.2ng/g.The analysis of the spiked samples showed that the established pretreatment and LC-MS methods could be applied to analysis four nitrofuran metabolites and malachite green residues in these tissues. The results have highly reliability.
