Investigators pay more attion to the problems of environmental security along with widely used of chiral pesticides.So we take human neuroblast cells(SH-SY5Y)and zebrafish(Danio rerio)embryo as models to study neuronal and developmental toxicity.The research can provide some new scientific basis for environmental security assessment and mechanism study.The detailed information is as follows:After the enantiomeric separation of mamathion by HPLC,the SH-SY5Y cells were treated by 10-7-10-4mol/L malathion and R-and S-malathion enantiomers respectively.The cell growth was measured by MTT,and cell morphology was observed by Microscope.The results showed that the growth of SH-SY5Y cells was obviously inhibited by rac-,R- and S-malathion with dose and time fashion.10-4mol/L malathion and its enantiomers displayed the most notable inhibition,and at that concentration the R-malathion inhibited the cell growth 2 times stronger than S-malathion after the treatment of 48 h,the relative inhibition ratio was about 75%.Meanwhile the results showed by microscope that cell morphology has been certain changed,such as deformation and size increasing.We also take zebrafish embryo as a model to study the influence of malathion and its enantiomers on development of zebrafish embryo.Confect malathion,R-malathion, S-malathion liquor of four different concentration gradient(10-6-10-3 mol/L)on zebrafish embryo to study the influence of development of zebrafish by morphological development,livability and percentage of hatching.The results showed that 6×10-5mol/L malathion and its enantiomers displayed obvious stereoselectivity on percentage of hatching,livability and lucity of zebrafish embryo.R-malathion showed the effect stronger than Rac-malathion,and Rac-malathion showed the effect stronger than S-malathion.Meanwhile the main abnormality of zebrafish displayed by R-malathion is vitelline duct cyst,and S-malathion is pericardiac cyst,Rac-malathion is deformity of spinalcolumn.The results tell us that the growth of SH-SY5Y cells was obviously inhibited by malathion and its enantiomers and the results showed obvious stereoselectivity of cell toxicity.At the same time malathion displayed the same stereoselectivity on the development of amniote embryo.We also take zebrafish as a model to study the influence of four pesticides on development of zebrafish embryo by morphology and histology.Confect tebuconazole,diniconazole,metolachlor and atrazine liquor of four different concentrations(10-6,10-5,10-4,10-3mol/L)on zebrafish embryo to study the influence of development of zebrafish by morphological development,livability and percentage of hatching.These four pesticides of 10-3mol/L have negative effect of death,abnormality and antifeedant activity on zebrafish embryo and also have time- and dose-dependent effect.These four pesticides can make death and abnormality on zebrafish embryo,and the research can provide some new scientific basis for environtoxicology of pesticide.In a word,we take in vivo and in vitro models to study toxic effects of chiral pesticides.The results of the study may instruct filtration of highly active and safety configuration and also make sense of environmental security assessment.
