Coenzyme Q10(CoQ10)was a prenylated benzoquinone lipid,and it was also named ubiquinone.The CoQ10were found in microorganisms, plants,and the heart,kidney,liver of animals,where they functioned as redox carriers in mitochondria or in association with respiratory processes of electron transport chains.It was an antioxidant produced naturally.In this paper,we studied the optimization condition of CoQ10extraction,the effect of the supplementation of PHB,glycerol,Fe2+and Mg2+on the fermentation of CoQ10,and the purifying technology of CoQ10.A novel isolate named Sphingomonas sp.ZUTEO3 was utilized to produce CoQ10,and the synchronous saponification extraction method was applied to extract COQ10.Influences of several factors such as saponification temperature and time,organic solvent volume on CoQ10 extraction were studied.The optimal conditions of synchronous saponification extraction were as follows:temperature 90℃,time 30min and organic solvent volume 30mL.This method has predigested and optimized the traditional saponification method with nonloss,which has opened vast vista.Finally,the extracted product was further purified by the column containing silicagel,and separated by Thin-layer Chromatograph,then determined by Ultravilet Spectrameter, Chromogenic Reaction.The results showed that it was oxidated CoQ10.The coupled process of fermentation and extraction was evaluated in the CoQ10production by Sphingomonas sp.ZUTEO3.The optimal conditions were obtained as follows:2%soybean oil as the cell membrane permeability accelerant was added into the original culture and 30mL hexane as the extracting solvent was added after 30h culture.After going sequential coupled process of fermentation and extraction for about 8h,the maximal yield of CoQ10could reach to 32.51mg/L.PHB is an important precursor in the biosynthesis of CoQ10.The optimal condition for the bioconservation from PHB to CoQ10were as follows:after 6h culture of Sphingomonas sp.ZUTEO3 in the optimal medium,15mg/L of PHB was added using distilled water as the solvent, and continued to ferment for 18h of biotransformationl.The maximal yield of CoQ10could reach to 69.91mg/L.The effects of the supplementation of glycerol,Fe2+and Mg2+on the yield of CoQ10were also studied.Through many experiments,When 1%glycerol,2mmol/L Fe2+,1mmol/L Mg2+was respectively added into the optimal medium,the yield of CoQ10could evidently increase.
