| We mainly research a new kind of aqueous two-phase separation system which is salts and low-molecular-weight organic solvents two-phase system. The studys refer to the basic phase-formed principles and the phase-formed time and the equilibrium of this system. We found the ammonium sulfate / alcohol two-phase system's effect was obvious superior to other systems in our research area. The mainly content as following:(1) We established a preliminary experiments and judge methods which used K salt and K organic solvents as a basis criterion to judge the capacity of the salts and the organic solvents. The method is better than the method which has been reported in the thesis of others. The phase-formed capacity of the salts and the organic solvents as following: (NH4)2SO4﹥K2HPO4﹥NaH2PO4 and ethanol﹥isopropanol.(2) We preliminaryly discussed phase-formed principles and the laws though researching on the phase-formed time of these systems.(3) Though the experiments we got the phase-formed concentration range of these systems upon 20℃as following: ammonium sulfate /isopropanol two-phase system isopropanol 11.02%~80.657%, water 18.684%~64.473%, ammonium sulfate 0.38%~38.87%; ethanol/ ammonium sulfate two-phase system, ethanol 7.077%~77.499%, water 22.353%~60.384%, ammonium sulfate 0.15%~37.82%; ethanol/K2HPO4 two-phase system ethanol 3.92%, water30.199%, K2HPO4 0.68%; ethanol/NaH2PO4 two-phase system ethanol 7.936%~71.934%, water 22.146%~59.038%, NaH2PO4 0.138%~55.50%.(4) Through experiments we eventually determined ethanol / ammonium sulfate two-phase system to be the best extraction system.(5) We established a new kind of method to inspect the affecting factors such as component content and temperature etc how to affect the distribution of rutin. Finally we found the optimum conditions: ammonium sulfate 25%(wt%) ethanol 22%(wt%) water 53%(wt%); extraction temperature 20℃~30℃.(6)We established two separate processes and found using aqueous two-phase systems to refine the early products will greatly enhance the content of flavonoids. |
