The total alkaloids from embryo of the seed of nelumbo nucifera gaertn include liensinine neferine, isolisensinine, and so on. They have activity of cardiovascular system. It is proved that have the functions such as lowering blood pressure, resisiting arrhythmia. Methods which the total alkaloids are extracted from embryo of the seed of nelumbo nucifera gaertn include traditional crafts such as impregnant extraction and new technologies such as film separation. Moreover, it's first time to use the macroporous adsorption resin to extract the total alkloids. The macroporous adsorption resin is a kind of macromolecule adsorbent which doesn't contain exchange groups but macroporous structure. It has great capacity to adsorption because of specific surface area, good selectivity to adsorptive substance, rapid adsorption and easy separation from itself. The physical and chemical characters of the macroporous adsorption resin are stable. It can be used for a long time, and cost saving due to using again and again.Free radical is a kind of live molecule. It can be produced by the human body's activity. But excessive free radicals inside the human body can cause cell and organization organ hurt, result in various disease and accelerate body decrepitude. Therefore, it's necessary to look for good antioxidant.In this thesis ethanol was chosen to carry on the alkaloids from embryo of the seed of nelumbo nucifera gaertn. Through the investigation of single factor experiments and influences of ethanol concentration, volume, circumfluence extraction time, an appropriate process is selected by orthogonal test. It is obtained that better route of circumfluence extraction is: extracting twice, 90% ethanol which use ratio 1:10 of material quality and ethanol volume, and extracting 60 mins for the first time, 90% ethanol which use ratio 1:10 of material quality and ethanol volume, and extracting 45 mins for the second time. Among them, the ethanol volume shows significently influence towards extraction.The adsorptive and desorptive experiments were performed after obtained the liquid of alkaloids. Firstly, the resin type was optimized for purification of the alkaloids from embryo of the seed of nelumbo nucifera gaertn. Through the investigation that adsorptive capacity in static state, ultimately, DA201 resin was chose to use. Through research on dynamics and thermodynamics characteristic, dynamics curves and thermodynamics data are concluded. It is proved that adsorptive process with freeing heat to DA201 adsorbed alkaloids is rapid and self-moving. The results of the orthogonal test of factors havae indcated that the optimum conditions are as following: (1)absorption conditions: concentration 1.87mg/ml, pH 10, volume 8BV , velocity of flow 2ml/min, the most adsorptive capacity is 14.02mg/g; (2)desorption conditions: ethanol concentration 70%, pH 3 , volume 8BV, velocity of flow 2ml/min. the first desoptive ratio is 49.17%. After desorbed three time, desorption ratio is 76.09%. Finally, the separate ratio is 40.53%.Anti-oxdation for the total alkaloids from embryo of the seed of nelumbo nucifera gaertn was studied by differerntial pulse voltammetry. During 0~0.5V, it is found that the redox peaks current at 0.27V increase with concentration of the total alkaloids. This provides a new method for measuring concentration of the total alkaloids from embryo of the seed of nelumbo nucifera gaertn. The study on scavenging effect extraction on superoxide anion and hydroxyl free radical suggested anti-oxidation of the total alkaloids from embryo of the seed of nelumbo nucifera gaertn is better than vitamin C. When the total alkaloids concentration is 1.0mg/mL, the ratio of scavenging on OH·is up to 90%, on O2ˉ·to 98%. It is proved that the total alkaloids have strong anti-oxdation.
