Study And Fabrication On New Nerve Conduits With Braided Structure And Working On It's Properties

At present,many scholars have made a great deal of research on the braided nerve conduit,not only elected the appropriate materials and braiding structure,but also did a lot of study on the performance of nerve conduit.However,nowadays nerve conduit can not meet the clinical demand.In cell and animal experiments,as the single nerve conduit is not beneficial to cell adhesion and growth, and can not provide carrier for nerve growth.The structure is not in favor of nerve growing to the far ends,which affects the speed and quality of nerve repair.As a result,the study focus on the inner layer of nerve conduit to make new braided nerve conduit.The new nerve conduit can improve the quality,the speed and the distance of nerve repair.This task has been developed the process of nerve conduit on the basis of ever braided nerve conduit.By optimizing the knitting process and parameters,produce more fitful braid-biodegradable nerve conduit. Through a series of performance study,which includes the pore rate, mechanical properties,out body degradation experiments,PH value measurement and cell cultivation experiments researching the new type nerve conduit.And discuss the new nerve conduit structure's infection on the cell cultivation and the nerve growth.Through looking for information and experimental studies,we determine the three production of new nerve conduit.The three type new nerve conduits are bonding parallel yarn as inner nerve conduit, fluffy parallel yarn as inner nerve conduit and chitin non-woven fabric as inner nerve conduit.Bonding parallel yarn and fluffy parallel yarn are made of PGLA fiber.Chitin non-woven fabric is not only with good biological performance,but also has similar degradation rate to PGLA.Experiment results can be shown that the three new braided nerve conduit have the similar pore rate with single-layer braided nerve conduit,but mechanical properties are better than single-layer one.In order to let new type conduit have a better performance,each type prepare three projects,compare properties and determine the optimal knitting method.In order to prove the superiority of the new nerve conduit,We have commissioned the Shanghai First People's Hospital to do cell planting experiment.This cooperation,not only to fully verify the new nerve conduits inner layers have good compatibility and non-toxic,but also this is the first time successfully plant the umbilical cord mesenchymal stem cells in braided nerve conduit.To analyze the results furthermore,we know that the chitin non-woven fabric can improve the cell adhesion.Chitosan coating on bonding parallel yarn,not only provide the carrier channel,but also increase nerve conduit cell compatibility.Fluffy parallel yam structure is well to three-dimensional structure for umbilical mesenchymal stem cells and separate propagation.Meanwhile,gaps in the parallel yam is conducive to regenerate axon adhere and extend.The new type nerve conduits in this study,is the first braided nerve conduit of intemal support structure in china.The reasonable three-dimensional structure makes the outer layer has good permeability,which in favor of the exchange between inner and outer.The inner layer for provide carriers for cell,which is well to three-dimensional culture for umbilical mesenchymal stem cells and separate propagation,and beneficial to cell adhere and axon extend. This study makes a contribution for the braided nerve conduit development.
