Study On The Composition And Activity Of Polysaccharides From Panax Japonicus C.A.Mey | | Posted on:2010-09-24 | Degree:Master | Type:Thesis | | Country:China | Candidate:B Xu | Full Text:PDF | | GTID:2121360278451754 | Subject:Applied Chemistry | | Abstract/Summary: | PDF Full Text Request | | On the basis of systematic research of extraction,isolation,purification process,physiochemical properties and chemistrical structure were studied on Panax-japonicus C.A.Mey polysaccharides , a clear chemistrical structure of the homogeneity polysaccharide from Panax-japonicus C.A.Mey was obtained,whose activity was systematically studied and evaluated.The mian contents and conclusions are listed as follows:1.Deionized water method and enzyme method were respectively used for extraction process of polysaccharides constituents from Panax-japonicus C.A.Mey,which were compared from the yield of polysaccharide.The research results indicated the yield of polysaccharide was more by using enzyme method and effectively zymohydrolysis cellulose and pectines through optimized process.The enzyme extraction process was simple, the extracted time was short,the yield of polysaccharide was much and did not break the structure.The yield of polysaccharide from Panax-japonicus C.A.Mey was 23.2% through enzyme method and was 11.61% through deionized water method.Obviously,the yield of polysaccharide form Panax japonicus C.AMey could be efficetively increased by using enzyme extraction process.2 . To study on the removal of protein in the polysaccharides from Panax-japonicus C.A.Mey. Sevag method,TCA method and caroid method were chosen . Comparing with effects of the removed protein and the recoveried polysaccharide,the yield of removal protein and the rate of recovery polysaccharides were used to evaluate the factor levels.The results indicated that the yield of removal protein and the rate of recovery polysaccharides were highest in caroid method.The conclusion was that caroid method can effectively remove protein in the Polysaccharides from Panax-japonicus C.A.Mey.3.Adopting active carbon and resin to decolourize the extracted polysaccharide,it was fund that the decolour effect of D101 resin was better than that of active carbon. The absorbance of polysaccharide by active carbon cause the polysaccharide content dropped. For D101 resin,there is not such phenomenon. D101 resin was a ideal decolournat.4.There are the homogeneity polysaccharides from Panax-japonicus C.A.Mey through DEAE-cellulose ion exchange resin and glucosan gel Sephadex G-75,which did not have nucleic acid and protein by ultraviolet scanning. It was indicated by FTIR that the homogeneity polysaccharide was a neutral polysaccharide, which was mainly composed of pyranose without alduronic acid.Through monosaccharide composition analysis, the homogeneity polysaccharide was composed of glucose and galactose. By HPGPC, Mp of the homogeneity polysaccharide was 6383. The research of the homogeneity polysaccharide had not been seen from the related research reports.5.The activity of homogeneity polysaccharide (PJCP-1) was evuluated.It was suggested by animal experiment that the polysaccharide PJCP-1 from Panax-japonicus C.A.Mey could be anti-aging effects.It could resist the atrophy of thymus and spleen of model mice,maleic dialdehyde levels in blood were decreased dramatically on model mice,superoxide dismutase,glutathione peroxidase and catalase activities in blood,liver and brain were increased obviously.By removing excess free radical and breaking lipid oxidation in the body,the Panax-japonicus C.A.Mey polysaccharide has anti-aging effects by immune-regulatory and eliminating free radicals. | | Keywords/Search Tags: | polysaccharide of Panax-japonicus C.A.Mey, abstraction, isolation and purification, activity evaluation | PDF Full Text Request | Related items |
