The present study was to take capsicum peel as the research materials. The research had investigated the extraction and purification and decolorization of capsaicinoids. And the inhibition of capsaicinoids to several microbes was preliminary studied in this paper.First, the research had compared the extracted results of using organic solvent extraction, using microware and ultrasonic. The optimum extraction conditions of organic solvent extraction were as follows: extraction solvent was 70% ethanol, ratio of material and liquid was 1:15, extraction temperature was 60℃, and extraction time was 100 minutes. The optimum extraction conditions of microwave radiation extraction were as follows: extraction solvent was 75% ethanol, microwave output power was 400W, ratio of material and liquid was 1:20, and extraction radiation time was 3.75 minutes. The optimum extraction conditions of ultrasonic radiation extraction were as follows: extraction solvent was 75% ethanol, ultrasonic output power was 350W, ratio of material and liquid was 1:15, extraction time was 10 minutes, and ultrasonic radiation time was 6 seconds. Microwave radiation extraction was better than ultrasonic radiation extraction and organic solvent extraction.Second, the research had compared the effect of extraction technology of preparing capsaicinoids. In the four extraction process, the results showed that the extraction technology of preparing capsaicinoids was ethanol extraction, extraction with ethyl acetate after concentration.According to the above technology, the purity rate of capsaicinoids of capsicol was 6.28% and the color value of it was 28.65. However, the purity rate of the capsaicinoids of capsicol extracted by Chenguang Company was 4.03% and the color value was 177.82%. Compared with the latter, the former is beyond 2 percentage points of purity rate, and the color value was little. Therefore, it was a new way of preparing capsicol and has high practical value.Third, the research had investigated the purification and decolorization of capsaicinoids. The results showed that the resin was NKA-9, absorption time 6 hours, absorption pH was 7, the exemplar concentration of 1.51g / L. The solvent was ethanol and its concentration was 80%, the pH of solvent was 5, absorption flow was 2 BV/h, desorption flow was 1 BV/h. The decolorization rate of capsicol was 87.36 %. On this basis, further study of capsaicinoids was crystallization conditions. The solvent of crystallization was ether, the temperature of crystallization was -5℃, the time of crystallization was 2 hours. Under this process, the purity of capsaicinoids was 93.4%.Finally, the inhibition of capsaicinoids to several microbes was studied. The results showed that capsaicinoids has inhibition. The best inhibition was to yeast. But to bacteria and streptomyces, the capsaicinoids has no inhibition. The MIC required different concentration of capsaicinoids inhibiting several microbes were as follows. Rhodotorula glutinis was 80 mg/L, Debaryomyces hansenii was 120 mg/L, Cryptococcus laurentii was 240 mg/L. The pH and temperature affected capsaicinoids antimicrobial action a little. |
