Capsaicin is a kind of bioactive chemical that is widely used. In this work, firstly, we primarily optimized the conditions using single-factor tests, and according to that we designed an orthogonal test with recovery of capsaicinoids and time interval as two indicators were performed to get the best extraction conditions through 95% ethanol. And the best conditions were selected according to Integrated Balance Method:extraction cycle was 20, extraction temperature was 140℃, volume of solvent(95%ethanol) was 130 mL and weight of pepper powder was 20g respectively. The recovery of capsaicin was 9.813mg/g and time interval between siphons was 7min, so total time used was 140min. Secondly, acid-base extraction method was used to separate capsaicin from capsaicinoids. And the optimized conditions were:the weight concentration of NaOH was 15%, Solid-liquid ratio 15, temperature 60℃, saponification time 1.5h, pH adjusted back to about 7.0. At last, the re-crystallization influnce of petroleum ether, ethyl acetate, acetone and ether was inspected. The results showed that ether was the best crystallization solvent for capsaicin, and the infrared method was used for analysis.After that, coupled reactor/column system was used to study the processing of n-butyl acetate with n-butanol and acetic acid. Aspen plus was used to found, analogue and optimize the process for esterification. The results were concentration of n-butyl acetate was approximate to 1.000 and conversion rate was about 0.987. On this basis, the recovery of acetic acid from variously waste water was performed. The result indicated that coupled reactor/column system was a highly effective way to manufacture n-butyl acetate with high purity and to recycle back acetic acid of various concentrations. Conversion rate of diluted acetic acid was enhanced, while energy cost decreased as the concentration of acetic acid increased. This study provided a useful reference to industrial processing.
