Coumarin derivatives are of interest because of their bactericidal, anticoagulant and anti-HIV activities.3 or 4-substituted hydroxycoumarins are used as active,non-peptide HIV protease inhibitors.4-hydroxycoumarin is considered to play a major role in the structural parts. For this reason, the optical and chromatographic propertities of novel derivatives, together with the development of new synthetic methods, are topics of growing interest for a great number of research groups.The synthetic progresses of coumarin derivatives were reviewed. Furthermore, a series of coumarin derivatives were synthesized under microwave-one-pot and their optical and chromatographic propertities were studied. These studies would provide useful informations and datas for the further exploitation of medicinal resources. The main contents are as follows:1. The recent advances in the synthetic methods of coumarin derivatives,advances of organic fluorescent dyes and HPLC applications in the analysis of coumarin derivatives were reviewed. The synthesis of coumarin derivatives were introduced.2. Four kides of (1,4)-bis-phenylenebis(methanetriyl)-[bis-(4-hydroxy-3-coumarin) ](a),(3,4)-bis-(4-hydroxy-3-coumarin)-2,5-hexylene glycol(b),(2,3)-bis-(4-hydroxy-3-coumarin)-1,4-Naphthoquinone(c),4-methyl-7-hydroxycoumarin(d) had been fast synthesized under microwave-one-pot. Reaction time was reduced and the yield was increased. Also, the microwave power,temperature and time on yield were investigated and the best synthesis conditions were explored. Their structures were verified by IR,1HNMR,13CNMR.3. The fluorescence and UV properties of aimed compounds were preliminarily studied. The results showed that powered group-hydroxy,rigid "flat" of 4-methyl-7-hyd roxycoumarin had strong fluorescence. The fluorescence intensity increased with increasing polarity; The fluorescence intensity increased with increasing of polarity in dioxane-water binary solvents; In the range of pH 3.5 to pH 5.1, the fluorescence intensity declined with declining of pH and in the range of pH 6.47 to pH 6.98, the fluorescence intensity declined with increasing of pH; In the temperature of 8℃to 20℃, the fluorescence intensity declined with increasing of temperature.4. Chromatographic behaviors of aimed compounds were studied preliminarily. Samples were detected using Kromasil C18 column. Operation conditions were set as follows:mobile phases V(acetonitrile):V(0.85%phosphoric acid)=50:50, column temperature 30℃,flow rate l.OmL/min, detection at 310nm. Results showed that good linear relationship were within 5μL~20μL with correlation (r) 0.999. Relative standard deviation (RSD) of peak area were both below 3.5% in five consecutive tests or repeated tests during 12h. The method is simple,rapid and accurate, which can be used for detection of drugs.
