Fuzhou West Lake is a scenic city park lakes, as recent industrial and agricultural production and human interaction that increase in magnitude of lake eutrophication. The lake microbial biosphere of material cycling and energy flow of the main participants in the lake ecosystem plays an important role.The paper-based 16S rDNA of PCR-DGGE (denaturing gradient gel electrophoresis) Microbial diversity of the West Lake was analyzed and studied. SDS-proteinase K method with water samples of the total bacterial DNA, and were separately purified 16S rDNA V3 and V6 V9 areas of PCR, then 20% -80% by denaturing gradient gel electrophoresis (DDGE) separation, the gel imaging systems to observe the sample DGGE denaturing gradient gel bands, different types of bacteria DGGE bands observed in different positions. The results show that horizontal and vertical sections in the direction of some differences in bacterial diversity. Central longitudinal 10cm deep water lake water bacterial diversity than 50cm of water depth; and farther away from the discharge port side vertical section office, 10cm and 50cm deep in water depths of water, the diversity of microbes little difference, to reflect the microbial populations in water diversity.This study will help us better grasp the distribution of microorganisms Lake lake and its lake ecosystems function and effect on lake depth to improve the ecological environment of great significance.
