| Broussonetia(Broussonetia papyrifera (Linn.) Vent.)is an important ecological and economic forest tree species in Chain, the most abundant resources. This article studied extraction technology for Broussonetia leaves the chlorophyll and protein, and the amino acid composition of Broussonetia leaves leaf proteins were analyzed, provided theoretical basis, for the comprehensive development and utilization of the Broussonetia leaves resources.The results are as follows:1. By the single factor which influence the ultrasonic assisted extraction of Broussonetia leaves chlorophyll extraction process parameters:temperature, extraction time, ultrasonic power, liquid-solid ratio, etc. The optimum condition was determined by the response surface analysis methods, which were: temperature 60℃, extraction time 50min, ultrasonic power 500W, liquid to solid ratio of 10ml/g.Under these extraction condition, the extraction rate of Broussonetia leaves chlorophyll reached 1.28%.2. By the single factor test, which influence the extraction of Broussonetia leaves protein by Alkali-soluble extraction process parameters:extraction temperature, extraction time, pH, liquid-solid ratio, etc. The optimum condition was determined by the response surface analysis methods, which were: extraction temperature 50℃, extraction time of 70min, pH9.0, ratio of liquid to solid 40ml/g. Under these extraction condition, the extraction rate of Broussonetia leaves protein reached protein 14.32%.3. The amino acid composition of Broussonetia leaves protein was analysised using automatic amino acid analyzer. The result showed:the complete hydrolyzate of Broussonetia leaves protein contained 18 amino acids. The content of the total amino acids was 822.5mg/g. and essential amino acids was 277.5mg/g,which was 33.7%of the total amino acids, the semi-essential amino acidsand the flavor amino acidswere 4.1%and 16.0%of total amino acids. |
