Clarifying the Vero cells deformation mechanism in the non-polar organic pollutants stress, and establishing a quantitative characterization of cell deformation method of markers on this basis will provide a theoretical basis of quantitative evaluation methods and technical support for the establishment of the deformation of biological safety of drinking water-related materials.We studied the cause of Vero cells'state affected by typical organic pollutant with the method of Flow Cytometry on the basis of the impact of several typical polar and non-polar organic pollutants. The results show that the organic pollutant leads to the plasma damage before the cell proliferation and death. And cell deformation may be the macroscopic performance characteristic of membrane damage.Then we extract Vero cells'membrane with the method of two-phase distribution.In this study, the best extraction conditions of plasma membrane were 6.6% (W/W) substrate concentration, salt concentration of 10 mmol.L-1. At this point Vero cell plasma membrane recovery is up to about 70% of high purity. Further study show that membrane protein and phospholipids accounted for the proportion of dry cell weight in organic pollutant is lower than those in normal cells. And the higher the concentration of organic pollutant is, the lower the proportion.This is a preliminary indication that Vero cell shape changes and membrane damage are related to the stress of non-polar organic pollutants. Phospholipids and plasma membrane protein content and percentage of change in cell membrane may lead to the changes of rigid structure and the deformation of cells. Therefore, phospholipids and plasma membrane protein content and percentage of change in cell membrane may characterize the deformation rate of Vero cells.
