β-cyclodextrin (β-CD) inclusion method is a convenient,quick chemical method to remove cholesterol from butter. This paper mainly studies the extraction technology of cholesterol in freeβ-CD and immobilizedβ-CD inclusion removal butter, which including the research of cholesterol in freeβ-cyclodextrin inclusion removal butter, immobilization ofβ-CD, and cholesterol in immobilizedβ-cyclodextrin inclusion removal butter,which including the research of cholesterol in freeβ-cyclodextrin inclusion removal butter, immobilization ofβ-CD and the structure of immobilizedβ-CD complex, cholesterol in immobilizedβ-cyclodextrin inclusion removal butter, and the structure ofβ-CD cholesterol inclusions.In this paper, the two enzymes method (total cholesterol kit) was used to determine the content of cholesterol in butter, the result shows: using alkalescent alcohol solution saponification 65℃/ 40min, N-Hexane for extraction, then use total cholesterol kit for determination, we can get that the content of cholesterol in butter is 299.8mg/100g, the recovery rate is 99.50%. In this study, we use the single factor experiment and the orthogonal experiment to confirm optimal process to remove the cholesterol in butter underβ-cyclodextrin inclusion method: add 3 times ofβ-cyclodextrin in butter, stir for 30min under 60℃, then let stand for 30min under 4℃, washing, centrifuge for 10min at 2000r/min, the removal rate of cholesterol can achieve 90.52%,β-cyclodextrins retention rate is 2.80%, it indicates that this process has high cholesterol removal efficiency.With chitosan for the carrier, using physical adulterating method to immobilizeβ-cyclodextrin(β-CD). Optimized immobilized conditions are as follows: the addition amount ofβ-CD and chitosan is 4.00 g / 100ml and 3.00 g / 100ml, respectively. And stirring time is 45min, the concentration of NaOH solution is 1.00 mol/L , leave it alone for 1h. Using the process mentioned above, the immobilized quantity can achieve 0.34gβ- CD/g chitosan.Using immobilizedβ-CD inclusion method to remove cholesterol in butter and single factor experiment to determine optimized conditions: water content of immobilizedβ-CD is 70%, the addition amount of immobilizedβ-CD is 2.4g /g butter, the ratio between butter and water is 1: 20, under this condition, the removal rate of cholesterol in butter is above 95% . Then use twice revolve combination experiment to build the mathematical model of removing cholesterol by immobilizedβ-CD in the butter.Exploring the optimal process and conditions to remove cholesterol from immobilizedβ-CD cholesterol compound: the concentration of ethanol solution is 95%, pH value is 10.0, reflow 1h and the temperature is 80℃, under this condition, the cholesterol removal rate is 92.91%.
