During the making process of Dao Huaxiang-flavor liquor,the formation and the changes of microflora decide metabolisms way of materias and energy in Daqu,also to a large extent decide the formation of special trains of Dao Huaxiang-flavor liquor.In this research,we examined dynamic changes of microflora and dominant microbes in Dao Huaxiang Daqu by using traditional identify methods and DGGE, clone analysis, etc. modern molecular biology methods.The results showed us:(1)Analyze Dao Huaxiang Daqu during four different periods by using traditional plate cultivation and classified statistic,the result showed us Daqu conclude four mainly microbial communities , respectively bacterium,yeast,mould and actinomycetes.The number of bacterium and yeast decreased and then recovered slightly.Thhe number of mould and actinomycetes decreased gradually.(2)Analyze the physiochemical indexes of Daqu during four differentperiods,include water,acidity,liquefying power,esterifying power and crude starch.The result showed us the changes of physiochemical indexes and microbial communities display certain rules.And the changes of microbes lead to the changes of physiochemical indexes.(3)PCR-DGGE analysis based on 16SrDNA was used to study the changes of prokaryotic microbial communities in Daqu during four different periods,16SrDNA fragments was amplified by using universal primers(341fGC/518R)from total DNA.Then separate the amplified 16SrDNA by DGGE,Through sequencing the dominant lines ,analysis of gene sequencing and Phylogenetic tree can be done.The result showed us prokaryotic microbes in Daqu had more rich diversity exsisted obvious dominant community structure change. Mainly include bacterium and actinomycetes, bacterium focused on bacillus and coccus takes majority,bacillus had more quantities and species.The kinds of actinomycetes were more single ,take Streptomyces as major.(4)PCR-DGGE and phylogenetic analysis based on 18SrDNA were used to study the changes of eukaryotic microbial communities in Daqu during four different periods, 18SrDNA fragments was amplified by using universal primers(NS-GC/EF4)from total DNA.Then separate the amplified 18SrDNA by DGGE,Through sequencing the dominant lines ,analysis of gene sequencing and Phylogenetic tree can be done.The result showed us eukaryotic microbes exsisted obvious dominant community structure,mainly lnclude yeast and mould,both had rich diversity,mould takes Aspergillus as major.The changes of superior communities and communities structures display a certain dynamic rules.
