Organophosphorus pesticide is one of most important chemical pesticides. Chorpyrifos as a kind of organophosphorus pesticides is high efficient broad-spectrum used in agriculture. It is a big problem that the chorpyrifos make the pollution with the food and enviroment. The principle of acetylcholine which makes the organisms been inhibited is a big problem.The Enzymic degradation was studied for the pollution of Ops, chorpyrifos using Pseudomonas stutzeri JHY01 strain as degradation bacteria. The present research was focusing on:the extraction methods of intracellular bacterial enzymes, the formula and stability of the organophosphorus pesticides liquid enzyme preparation, evaluating its degrading effect. The results are as follows:1) Extracting the intracellular enzymes of JHY01 strain:it been proved by research that ultrasonic fragmentation method is more efficient than bead mill method and chemical osmosis.The protein content of crude enzyme is 14.9g/L; chemical osmosis method is 12.84 g/L, bead mill method is 0.128g/L. The enzyme activity of chemical osmosis is 73.05%. So, the chemical osmosis is suitable method.2) Effect and injury repair of chorpyrifos for cucumber seedlings:proved by bioassay,chorpyrifos has influence to the plant height, fresh weight, leaf weight, stem length, leaf area, root (taproot length, lateral root length and lateral root number). In the range of 0-80mg/kg, the higher concentration, the inhibition rate of growing, chorpyrifos has higher influence to the root length and leaf area.,and EC50 values were 13.5 mg/kg and 16 mg/kg, repairing the cucumber seedlings by crude enzyme has good effect.3) Preparation and determination of stability with Organophosphorus pesticides liquid enzyme preparation:filtering CAN,NYG,DUBE,EROL and OSTC which can protect the enzyme.Research fixed the content of protective substances by determining the degradation of single protective substance. Degradation rate of crude enzyme is stable during 72.81%-84.89%. The research also fixed the formulation and content of protective substances in 7 days. The results proved that the enzyme preparation can't lose its activity between 25℃and 40℃and also can keep its degradation; the light isn't significant effect to degradation rate of crude enzyme. Liquid enzyme preparation can keep the activity as 61.15% in 3 months.4) Evaluation of degradating effect by liquid enzyme preparations:it was detected the chorpyrifos residue on the cucumber seedlings and in soil by HPLC. The result improved that the degradation rate was 88.71% in soil during, and was 72.61% during twelve hours and 77.46% during one day on the cucumber seedlings.
