| In recent years, the water eutrophication is a serious environmental problem.And eutrophication produces one of the most important problem is bcyanobacterial blooms.In nature, many species of algae can produce one or more cyanobacterial toxins, structure and toxic mechanism and toxicity of different toxins have very big difference. The microcystin algae toxin- LR(MC-LR) the most damage.At present, the degradation of algae and MC-LR methods can be divided into physical, chemical and biological methods.Microorganisms in biological methods for algae removal and degradation of toxins is a hot issue in recent years.The study found that white-rot fungus has good degradation effect, especially for refractory can better degradation of various pollutants, so this experiment studied the removal efficiency and mechanism of Microcysis aeruginosa and microcystin-LR by white-rot fungus.The experiment selected the four kinds of white-rot fungus :Phellinus pini,Piptoporus betulinus,Fomes fomentarius and Tremetes suaveloens.The test is divided into five parts :the cultivation of algae,the cultivation of white-rot fungus,white-rot fungus test to remove algae, the white-rot fungus remove the MC-LR experiment and two kinds of enzymes degrade the experiment of MC-LR.First, in the white-rot fungus remove algae experiment, we uses the determination chlorophyll a content the method to conduct the experiment.The test result showed that the white-rot fungus is best in the Kirk culture medium raises the elimination effect,and most suitable p H value of response is 7.55, the most suitable throwing increment is 10%-15%.Four kinds of white-rot fungus compared under the same condition,Phellinus pini’s elimination algae effect is best,removal efficiency is more than 60%.Second, in the degradation of microcystin-LR algae toxin white-rot fungus experiment, tested by high performance liquid chromatography(HPLC) method. The test result shows the special culture of white-rot fungus uses the liquid medium culture method, and the reaction system, the optimum p H value of 4.5, the optimum reaction temperature 30℃, reaction should be under static conditions.Four kinds of white-rot fungus compared under the same condition,Phellinus pini’s elimination algae effect is best,the degradation rates were above 85%.Finally, in the enzymatic degradation of MC- LR experiments, laccase has no obvious degradation effect, and manganese peroxidase has good degradation effect,the degradation rates were above 50%.In this article, through a different angle to study the removal efficiency and mechanism of Microcysis aeruginosa and microcystin-LR by white-rot fungus,to find the optimum conditions,understands the degeneration mechanism of white-rot fungus degeneration algae and algae toxin initially,to provide scientific basis for the future research work. |
