Cobalamin is one of important productions that is fermented in china. It is widely used in medicine and food industry. It is producted mainly by fermentation. The influence of several organs substrate propionic acid on Propionibacterium freudenreichii growth and VB12 fermentation were investigated. The fermentation process of removal propionic acid was also studied. The study was based on industry applications, which have special valuable reference to improvement of cobalamin fermentation.1. The influence of substrate on deoxyadenosylcobalamin fermentationThe effect of different glucose concentration and corn concentration on the Propionibacterium freudenreichii growth were investigated. The results showed that the glucose of 10.0 g/L-30.0 g/L is more suit for growth of cell. The corn of 30.0 g/L-40.0 g/L is more suit for growth of cell.2. The influence of propionic acid on deoxyadenosylcobalamin fermentationThe effect of propionic acid added in the medium on Propionibacterium freudenreichii growth were investigated and changing for fresh broth take propionic acid off in vitamin B12 (VB12) fermentation broth was also evaluated. The results showed that when the propionic acid concentration of adding was 0-0.5%, the cellμmax was 60% of ordinary fermentation and when adding 1.0% propionic acid, the cellμmax was 30% of ordinary fermentation. The results of replacing fresh broth resulted higher concentration of cell, compared to ordinary fermentation. The OD of cell improved 74.5%. The yield of deoxyadenosylcobalamin improved 40.7%.3. The research on coupling fermentation technicsThe influence of adsorbing by resin ZGA330 to take propionic acid off in vitamin B12 (VB12) fermentation was evaluated. The fermentation process of removal of propionic acid was also explored. It can efficiently relieve the inhibition of cell growth and realize the continuous grow of biomass. And the research results can be the foundation for the technology of continuous fermentation. 4. The influence of extracting technics by membrane on Propionibacterium freudenreichii growth.The influence of extracting technics by membrane on Propionibacterium freudenreichii growth was investigated. The results showed that the cell growth is not restrained when the cell concentration before upto 50.0% using by membrane extracting.
