Cross was made between wheat-Thinopyrum intermedium substitution lines and wheat cultivar Yangmai 10, and the immature embryos were cultured and the regenerated plants were induced to introgress the BYDV-resistant gene from alien substitution lines to wheat. BYDV resistance screening and cytology analysis were carried on among the resistance of regenerated plant offsprings and to get translocation lines and screen molecular markers that could track the BYDV-resistant gene. The BYDV-resistant materal will be used in the wheat breeding program, and molecular markers would facilitate the translocation lines with BYDV resistance screening effectively.i. Through genomic in situ hybridization (GISH) with Thinopyrum intermedium (E1E2St) genomic DNA as probe and Triticum astivum (ABD )genomic DNA as a blocker, it is demonstrated that the two lines 04Y5579-2 and 04Y5579-3 are translocation lines,which contain a Robertson translocation chromosome between wheat and Thinopyrum intermedium . Using rye genomic DNA as probe ,the result of GISH analysis showed that , translocation lines 04Y5579-2 contains rye 1RS chromosome.ii.
