Objective To investigate the expression and relationship GDM placentas HIF-1α and EPO with placental injuryMethod64cases of maternal GDM GDM group,64cases of poorly controlled blood sugar22cases (A group), good blood glucose control42patients (group B).Select the same period37cases of normal pregnant women as normal control group (group C).3groups take two placentas, one to be extracted RNA; another one placenta paraffin sections, the expression of HIF-la detect EPO protein expression in placental tissue, and HIF-lamRNA and EPOmRNA’s.Statistical analysis was performed on the two sets of data were observed in the expression levels of placental tissue HIF-la, EPO protein, HIF-lamRNA and EPO mRNA’s.Result1HIF-lashowed strong expression (brown staining) in the GDM group placentas, EPO in GDM placental trophoblast cells in tissues between cells, endothelial cells and villous stromal cells showed strong expression, HIF-la and EPO in placental tissues in the control group and the control group the same position but was weakly positive.2GDM placental HIF-la protein expression positive rate of85.9%(55/64), and the control group32.4%(12/37) compared statistically significant (P<0.05). EPO placenta in both groups are shown in comparison EPO protein expression was89.1%(57/64), and the control group35.1%(13/37) compared statistically significant (P<0.05).3GDM placental tissue HIF-lamRNA levels (0.88±0.12), compared with normal pregnancy placental tissue was significantly higher (0.47±0.11), the difference was statistically significant (P<0.05); GDM group placentas EPOmRNA levels were (0.93±0.27), significantly higher than (0.43±0.14), two groups, the difference was statistically significant (P<0.05)4A, B, group C positive expression rate of placental HIF-la protein were90.9%,73.8%,32.4%, EPO protein expression rate were100%,88.1%,35.1%, the difference between groups was statistically significance (P<0.05); A, B, C group placental HIF-lamRNA expression levels were (0.9±0.13),(0.7±0.12),(0.47±0.11), EPOmRNA expression levels were (0.96±0.23),(0.75±0.27),(0.43±0.14), the difference between the two groups was statistically significant (P<0.05)5GDM huge group of children with birth weight HIF-1α and EPO expression was positively correlated (r=0.662,0.475, P<0.05).Conclusion HIF-la and EPO have high levels of expression in GDM placentas and placental hypoxia more severe the higher the degree of injury level.
