Flow cytometry is a conventional measurement technique at this stage at abroad, whichis mature in method, reliable in technology and easy to handle. Our research in the field offlow cytometry starts late, and commercial flow cytometry products have not yet beenlaunched. With the emergence of new technologies and methods, flow cytometry isdeveloping rapidly and tends to automation and high efficiency direction. The liquid flowsystem module is one of the most important functional module in flow cytometry instrument,which includes monitoring on the pressure ratio of the sheath and sample liquid, self-cleaning flow system and its internal pressure and construction of the whole flow cytometryinstrument platform. Reasonable sample liquid and sheath liquid inlet velocity ratio isnecessary for the formation of single cell flow so that correct test results can be obtained.Therefore, research on the liquid flow system is of great significance in the development offlow cytometry instrument.This paper aims to research and realize the liquid flow system of a flow cytometryinstrument, establish a flow cytometer flow system platform to realize automatic cleaningand congestion eliminating and to simulate flow chamber inlet velocity ratio and doublesheath flow chamber flow field.The main work is as follows:1) Simulating the flow field characteristics inside the double sheath fluid chamber.Proposing and implementing a double sheath fluid flow chamber with a wide range of speedchanges and allow better stability, reducing the requirements on the control precision of thefluid flow system.2) Designing the self-cleaning flow device of flow cytometry system. The device canautomatically detect plugging and remove it, which makes operation more convenient.3) Designing and building the entire fluid flow system platform. Achieving pneumaticdisplacement injection process, controlling the inlet velocities of the sample liquid andsheath liquid by monitoring the air pressure inside the flow chamber using pressure sensor.Through monitoring the pipeline pressure, implementing the real time judgment of flowsystem needle blockage and the flow chamber plugging and remove them in time, whichprovides a reliable guarantee for the obtain of the correct test results.4) Designing hardware circuit of the fluid flow system, which includes design and debugging of the module of electromagnetic valve control, pressure control and abnormalitydetermination, and analog to digital conversion.5) Designing controlling software for the fluid flow system, designing the softwareinterface and debugging the control instructions using LabVIEW.
