ObjectiveTo investigate the cardioprotection effect of sufentanil pretreatment in myocardial ischemia reperfusion injury, and the role of autophagy pathway in it.Methods24 healthy male SD rats (200 ~ 250 g) randomly divided into 4 groups:Sham group:only opening chest without ligation of coronary artery,; Sham+ sufentanil group:sham operation with sufentanil pretreatment, I/R group:open chest with 30 min of ischemia and 120 min reperfusion of coronary artery,; I/R+SF group:30 min of ischemia and 120 min reperfusion of coronary artery with sufentanil pretreatment.; After 120 min reperfusion,2 ml blood withdraw from postcava. CK-MB and LDH levels were tested; Transmission electron microscopy was used to observe the morphological changes of myocardial cell. Western Blot method was used to detect LC3-Ⅱ/LC3-Ⅰ, Beclin 1 and Bcl 2 protein expression.ResultsCompared with I/R group, myocardial enzyme (CK-MB values) in I/R+SF group fell to 1577.6 U/L from 2538.9 U/L, the difference was statistically significant. (P< 0.05); Compared with the Sham group, LC3-Ⅱ protein expression in the IR groupe increased obviously, the Bcl-2 protein expression significantly reduced, the difference was statistically significant (P< 0.05); Compared with the Sham+SF group, LC3-Ⅱ protein expression the I/R+SF group was significantly higher, and the difference was statistically significant (P< 0.05); Compared with IR group, the Bcl-2 protein expression the I/R+SF group was significantly increased. (P< 0.05).ConclusionAutophagy is involved in the cardioprotection of sufentanil pretreatmengt in myocardial ischemia reperfusion injury.
