| Nano-technology as a rising technology attracted widely attention with rapidly developing. The quantum Confinement Effect, Size Effect and so on of QDs and large surface area/volume, physical properties of magnetic bead were used for separation and enrichment of cells, labeling and detection of protein, dosing and treatment of targeting by science researchers and becoming more and more important,lung cancer was a malignancy with high morbidity and mortality. Nowadays, lung cancer become the first tumor in China and made large threaten for life and healthy. Most patients of lung cancer which were hardly treated well and could not live for long time were diagnosed as later period tumor. Thus, the early diagnosis was rather important for improving the chance of cure and increasing their living time. These were really hard for the methods of early diagnosis and developed the research for tumor markers.Tumor markers were some special proteins in the patients of cancer. Circulating tumor cell was a kind of special tumor cell which was stem from cancer of primary invading into vessel. It can be moved by blood and invade into other places of body with forming new tumor. The quantity of circulating tumor cell with relation to the stages of tumor had clinic values with early diagnosis, monitoring condition of patients and selection of scheme for treating.This paper was about dectecting tumor markers on molecule level and CTC on cell level to explore new ways to early diagnosis for lung cancer.1. In our study, QDs which showed bright fluorescence under UV-light was prepared as immune-QDs test paper to qualitatively and semiquantitatively detect the tumor markers NSE and CEA of lung cancer. This test paper had great specificity on detecting tumor markers. The limit of detection of QDs-based test paper for NSE(2ng/m L) which were better than Colloidal Gold-based test paper can detect tumor markers convenience, rapidness, delicacy and mass with high value.2. However, the quantity of circulating tumor cell was few and hardly monitored.So immune magnetic bead was used for separating and enriching cells which were also labeled by fluorescence material to confirm their quantity. In our study, we captured limited CTC from large number cells and detecting the amount of CTC by electrochemical impedance and fluorescence intensity. This method had theadvantages of high sensitivity, great specificity and analgesia with good value for clinic. |
