Study On Immune Mechanism Of Cang Bai Ganzhiqing And Its Separated Components On Mycoplasma Urealytic Mice

Objective1. To observe the effect of Cangbaishiduqing and its subdivisions intervening the establishment of UU model in BALB/c mice.2. To probe the possible immunological regulation mechanism of Cangbaishiduqing and its subdivisions intervening UU model in BALB/c mice from Interleukin-2 (IL-2) level in serum, Iriterleukin-12 (IL-12) level and leukotriene C4(LTC4) level of vaginal mucosa tissue.MethodUU4 lyophilized serum strains were recovery, continuous cell culture, adjust the concentration of the mixture of 105CCU/ml until the third generation. Set it aside.BALB/c mices were randomly divided into six groups--group A (cangbaishiduqing group),group B(promoting blood circulation and urination with diuretics group), group C(clearing away heat and toxic materials group), group D(invigorate the spleen to damp elimination), group E(model group),group F(blank group).First, group A,B, C, D and E were established UU model adopting Estradiol pretreatment method. Secondly, different chinese herbal subdivisions was intervening in group A,B,C and D,no intervention in Group E for 7 days.Group F did not make any intervention. In the end, the vaginal secretions of each mice were cultivated UU. Each mice was detected IL-2 in blood, IL-12 and LTC4 in vaginal mucosa tissue by ELISA.Results1. The engraftment rate in groups were,Group A 87.5%,Group B 88.89%, Group C 100%,Group D 88.89%,Group E 100%, Group F 0%. The group of chinese herbal subdivisions was no different between Group E (P>0.05).2. The blood IL-2 average from high to low in turn were Group A (36.066 ±4.277),Group C (32.148±4.656),Group D (28.831±6.593),Group E (27.876 ±3.984), Group B (25.554±4.565), Group F (21.361±4.865). The values of blood IL-2 in Group A were higher than the values in group E and F(P<0.05). The values of blood IL-2 in group C and D were higher than the values in group E(P<0.05).3. The Vaginal mucosa tissue IL-12 average from high to low in turn were, Group E (12.471±2.547).Group C (11.034±2.988), Group B (10.128± 3.602), Group D (8.757±2.957), Group A (7.719±3.944), Group F (6.662±2.189). The values of IL-12 in group E were higher than the values in Group F (P<0.05).The values of IL-12 in group A and D were lower than the values in group E (P <0.05).4. The Vaginal mucosa tissue LTC4 average from high to low in turn were, Group E(225.325±25.209),Group A(214.152±81.743),Group D(201.641±56.112),Group B(181.045±103.605), Group F(160.483±39.788), Group C (158.622±116.962). The values of LTC4 in group E were higher than the values in Group F (P<0.05).Conclusions1. Cangbaishiduqing could raise the blood IL-2 level.2. Cangbaishiduqing and chinese herbal compound for nourishing spleen and eliminating dampness could decrease the IL-12 level of vaginal mucosa tissue.
