| Purpose: To observe the change of Gastric mucosa and hypothalamic SubstanceP (SP), serum gastrin (GAS), Hippocampal5-hydroxytryptamine(5-HT)and BDNFmRNAafter giving electro-acupuncture treatment at Gan Shu (BL18) in gastric ulcerrats with depression, and according to the changes in these indicators to explorethe mechanism of the electro-acupuncture on the treatment of depression gastriculcer.Material and method: After the open-field test, the rats were randomlydivided into4groups: normal (n=15),model(n=15),BL18group(n=15) andnon-acupoint group (n=15).All of the rats are modeled excluding the normal. BL18group electroacupuncture at BL18, non-acupoint group electroacupuncture at thepoints that one cm beside the belly button, acupuncture with6mm,electro-acupuncture treatment given voltage2V, density wave type, sparsely wave4Hz, dense wave15Hz, retaining for20min, one time for a day, continue for6days and a break of one day, two weeks of a course of treatment, a total of13days. During the experiment, observed the general condition,the result of theopen filed test. After the treatment, calculating the ulcer index, and usingELISA,RT-PCR and immunohistochemistry to detect the change of serumgastrin,gastric mucosa and hypothalamic SP, hippocampal5-HTandBDNFmRNA.Results: After modeling, vs the normal group the other groups changedobviously with the general condition. The result of the open filed testdemonstrated the success of the depression model. After the treatment, the ulcerindex showed that the effect of the electro-acupuncture BL18is markedly superiorto that model and non-acupoint (P<0.01). The immuno histochemical results found,after electro-acupuncture treatment at Gan Shu (BL18), compared with the modelgroup, the contents of SP in gastric mucosa and hypothalamic tissue weresignificantly reduced (P<0.01). Hippocampal5-HT content increased significantly (P<0.01). The Gas content in rats was significantly reduced(P<0.05).BDNFmRNA contain increased significantly(P<0.05).After comparisonbetween model control group and model group, the indicators have significance,and the Model is validated. The data analyzed using one-way ANOVA and LSD test.Conclusion: Electro-acupuncture treatment at Gan Shu (BL18) reduceing thecontent of SP in gastric mucosa and hypothalamic tissue, and reduceing thecontent of GAS so that stomach ulcers can be alleviated(stomach is alleviated).Electro-acupuncture treatment at Gan Shu (BL18) can increase the content ofhippocampal5-HTand BDNF, so that the depressive state to be eased. Therefore,electro-acupuncture treatment at Gan Shu (BL18) not only can alleviate gastriculcer, but also can ease the depressive state. |
