Objective:To observe the effect of compound Uncaria hypotensive tablet on blood pressure of SHR, thoracic aorta, renal artery morphology, to study the effect of compound Uncaria hypotensive tablet on vascular remodeling in SHR CRP, MCP-1, MMP-9, and provide a theoretical basis for the treatment and prevention of target organ damage in hypertensive patients.Methods:2412week old male SHR according to the blood pressure divided randomly into3groups, namely the spontaneously hypertensive rat model group (group SHR-M), compound Uncaria hypotensive tablet group (group SHR-G) and irbesartan in the western medicine group (SHR-D group),8rats in each group; the other8WKY rats as blank control group (WKY group). All the rats were administered1times, continuous intragastric administration for6weeks, WKY group and SHR-M group with10ml/kg of distilled water by gavage with distilled water; group SHR-G compound Uncaria hypotensive tablet prepared into suspension, according to kg D,10ml/kg439.77mg/gavage with distilled water; group SHR-D will Irbesartan Tablets preparation the suspension, according to kg D,10ml/kg13.74mg/gavage.The applicability of animal feeding started after1weeks, once a day, treatment for6weeks during the intervention, a fixed time each week to a blood pressure measurement. The tail cuff method by hand in the stomach before, first weeks after intragastric administration, second weeks, third weeks, fourth weeks, fifth weeks, sixth weeks at a fixed time point of blood pressure measurement, statistical average value of rats in each group at each time point of blood pressure. After6weeks, the rats were killed, serum was collected and the thoracic aorta, renal artery specimens, ELISA serum concentration of CPR, immunohistochemical detection of thoracic aortic MCP-1and the expression levels of MMP-9, HE staining was used to observe the morphological changes of aorta and renal artery. Results:(1) first weeks after treatment began at the end of the experiment, rat tail artery pressure in SHR-G group was lower than that of group SHR-M, but the effect is less than that in SHR-D group.(2) the concentration of serum CRP in SHR-G group was significantly lower than that in group SHR-M, also lower than the WKY group, the SHR-D group had no difference.(3) the average optical density of SHR-G group of thoracic aortic wall MCP-1, MMP-9value decreased, protein expression was significantly decreased, and the SHR-G group and the SHR-D group has no significant difference.(4) in group SHR-G and group SHR-D of thoracic aortic wall, outer membrane was improved than the SHR-M group, and SHR-G group to WKY group change.(5) in SHR-G group and SHR-D group renal artery wall, outer membrane was improved than the SHR-M group, and SHR-G group and SHR-D group had no significant difference.Conclusion:Compound Uncaria hypotensive tablet can lower blood pressure, may be related to the regulation of vascular remodeling; can reduce the SHR of thoracic aorta, renal artery vascular remodeling, the expression may be related to the decrease of serum concentration of inflammatory factor CRP and the down-regulation of MCP-1, SHR in thoracic aorta endothelial cells MMP-9protein level.
