| Objective:To investigate the effect of bushentongluofang on the expression of transforming growth factor-β1,connective tissue growth factor and the proliferation in rat glomerular mesangial cells cultivated under rat interleukin-1β,in oder to explore its possible mechanisms to defer renal fibrosis.Methods:1ã€Using plasm pharmacological to prepare drug-containing plasm of BSTLF and fosinopril;2ã€Promote the proliferation of rat glomerular mesangial cells through rat interleuin-1β;3ã€The rat glomerular mesangial cells divided into control group, model group, fosinopril group, high-dose groupã€middle-dose group and low-dose group of drug-containing plasm of BSTLF;4ã€MTT method:To detect the effects to the rat glomerular mesangial cells linesproli feration by drug-containing plasm of BSTLF;5〠RT-PCR method:To test the rat glomerular mesangial cells TGF-β1-〠CTGF mRNA expression in24h.Results:1ã€The cell proliferation by MTT and flow cytometry:Compared with the normal group,In vitro cultured glomerular mesangial cells were obviously proliferating under 2, the stimulation of rat interleukin-1β (P<0.01).Compared the model group,the fosinopril group and high,middle,low dose of bushentongluofang inhibited the proliferating of glomerular mesangial cells(P<0.05).Compared the fosinopril group, there is no difference with high,middle,low dose of bushentongluofang(P>0.05)3, CTGF. TGF-β1mRNA levels by RT-PCR in each cell group: Compared with the normal group, the expressions of CTGFã€TGF-β1mRNA were increased dramatically under the stimulation of rat interleukin-1β(p<0.01).Compared with the model group, the expression of CTGF-. TGF-β1mRNA of the fosinopril group〠high-dose group〠middle-dose group and low-dose group of Bushentongluo-fang significantly decreased(P<0.01), Compared with high-dose,middle-dose,low-dose of bushentongluofang,there is no difference between them(P>0.05). Acertain dose-dependent was not shown.Conclusions:1〠rIL-1βcan stimulate the proliferation of GMCs and increase the expressions of CTGF, TGF-β1, mRNA.2ã€The drug-containing plasm of BSTLF can inhibite the proliferating of GMCs.3ã€The drug-containing plasm of BSTLF can decrease the expression of CTGF TGF-β1mRNA, it is similar to Fosinopril.A certain dose-dependent was not shown.All the results show that BSTLF can inhibit fibrotic factor to express,and it useful for reduceing the development of NS. |
